Özet

Amaç: Bu çalışmanın amacı nöronal otofaji ile status epileptikusun ilşkisini araştırmak veklorokinin epilepsi ve otofaji üzerine etkilerini incelemektir.Yöntem: Kontrol grubu, pentametazolle başlatılan epilepsi grubu ve klorokinin ile öncütedavi edilen grup olmak üzere üç ayrı gruba ayrılan 24 erişkin erkek Wistar sıçanı kullanıldı.Enjeksiyondan sonra 2 saat içersinde sıçanların elektroensefalogramları ve davranışlarıkaydedildi ve gözlendi. Nöron kaybı seviye tayini için Nissl boyaması kullanıldı. Otofajiyansıması olarak ise mikrotübüle bağlı protein hafif zincir 3 (LC3) ve p62 seviyeleri ölçüldü.Sonuç: Nöbet yoğunluğu klorokinle tedavi grubuna göre pentametazol grubunda daha ağır vekontrol grubuna göre ise de daha da ağırdı (p

Abstract

Objective: The present study aim to investigate the relationship between neuronal autophagyand status epilepticus, and to probe the effects of chloroquine both on autophagy and epilepsy.Method: 24 adult male Wistar rats were randomly divided into three groups, named controlgroup, pentamethazol induced epilepsy group and chloroquine pretreatment group. Thebehavior and electroencephalogram of rats were observed and recorded within 2h afterinjection. The Nissl staining was used to detect the level of neuronal loss. And the levels ofmicrotubule-associated protein light chain 3 (LC3) and p62 was reflected to the expression ofautophagy.Results: Seizure intensity in the pentamethazol treatment group was more severe than that inthe chloroquine pretreatment group and the control group (P
