Effect of Short and Long Term Omega-3 Fatty Acids on Cold Stress Induced Neuronal Changes in Hippocampus of Wistar Rats

Özet
Giriş: Kaçınılmaz stress, öğrenme ve bellek bozuklukları ile birlikte davranış defisitleriyleortaya konur. Böylesi stress kaynaklı nöronal kayıp için mevcut ilaç tedavileri yetersiz etki veyan etkileri nedeniyle engellenmektedir.Amaç: Bu çalışma kısa ve uzun dönem omega-3 yağ asitlerinin Wistar sıçanlarınınhipokampüsünde soğuk stresi ile oluşturulan nöronal değişikliklerine etkisini karşılaştırmakiçin tasarlandı.Yöntem: Bu çalışma için beş gruba bölünen (n=5) toplam 30 erkek sıçan kullanıldı. GrupIde kontrol amacı ile sıçanlar ideal laboratuvar koşullarında tutuldular. Grup 2de sıçanlarsoğuk stresi oluşturulmak üzere soğuk suda yüzmeye zorlandılar. Grup 3te soğuk stresssonrası ağızdan gavaj ile tuzlu su verildi. Grup 4ve 5te soğuk stress sonrası ağızdan gavajyolu ile sırasıyla 60mg/Kg docosahexaenoic asid (DHA) ve icosapentaenoic asid (EPA)omega-3 yağ asitleri 2 hafta (kısa dönem modeli) ve 4 hafta (uzun dönem modeli) verildi.Çalışmanın tümü 60 günde tamamlandı. Ertesi gün kortizol ölçümü için kan testi yapılarakdeney hayvanları sakrifiye edildi. Beyinleri disseksiyonla histolojik çalışma için ayrıldı.Sonuçlar: Hipokampüsün CA1 bölgesinde mikrometre kullanılarak total nöron sayısıhistolojik kesitlerde boyanarak hesaplandı. Veriler kontrol grubu sıçanlarınkilerlekarşılaştırıldı. Sonuçlar uzun dönem omega-3 yağ asitleri ile tedavi olanların diğer gruplaraoranla istatistiksel olarak daha önemli olduğunu ortaya koydu.Yargı: Günlük yaşamımızda stress ile başa çıkabilmek için uzun dönem omega-3 yağ asitleriile tedavi, bellek üzerine ek bir geliştirici etkisi nedeniyle, önerilebilir.
Abstract
Background: Inescapable stress is associated with impairments in learning and memorymanifested by deficits behaviors. The current drug therapies for such stress induced neuronalloss are hindered due to poor efficacy and side effects.Objective: Present study was designed to compare the effect of short-term and long-termtreatment with omega-3 fatty acids on cold stress induced neuronal changes in hippocampusof Wistar rats.Methods: Total thirty male rats divided into five groups were used (n=6) for this study.Group I was control in which rats were kept under ideal laboratory conditions, Group II wasgiven cold water swim stress in which rats were forced to swim in the cold water. Group IIIwas given cold stress followed by oral gavages of saline. Group IV and Group V was givencold stress followed by oral gavages of omega-3 fatty acids 60mg/Kg of docosahexaenoicacid (DHA) and eicosapentaenoic acid (EPA) 2 weeks (short-term model) and 4 weeks ( longterm model) respectively. The complete study was carried out for 60 days. The next day bloodwas collected for cortisol assay and then the animals were sacrificed. Brains were dissectedout and processed for histological analysis.Results: The total number of neurons was calculated from stained histological section byusing micrometry in the CA1 region of the hippocampus. The data were compared with thoseof control rats. Results indicate that long term treatment with omega-3 fatty acids isstatistically significant when compared to the other groups.Conclusion: Long-term treatment with low doses of omega-3 fatty acids suggested to be usedas an adjuvant to improve memory and to combat stress in our day to day life.
