Chronic Mild Stress Induces Memory Deficit With Asymmetrical Damages in The Hippocampus in Rats

Özet

Giriş: Majör depressif bozukluklar (MDB) bellekte defisitlerle birlikte sıklıkla hipokampalhasarlara bağlıdır. İnsan beyninde sol-sağ asimetrisi yüzyıldan beri bilinmektedir. Kronikılımlı stress (KIS) protololleri deney sıçanlarında depresyon benzeri davranışları uyandırmakiçin oldukça geniş çapta kullanılmıştır. Ancak hipokampal hasarlanmada sol-sağ asimetrisi iledepresyon arasındaki ilişkiyi araştıran çok az sayıda çalışma vardır. Bu çalışmanın amacıKISin etkilerini hem hipokampus hem de öğrenme ve bellek üzerinde göstermektir.Yöntem: KIS modelinin depresyon-benzeri davranışlarını kontrol etmek üzere vücut ağırlığıve sükroz tercih ile modifiye bir açık-alan testi gerçekleştirildi. Hipokampusun öğrenme vebellek, apoptoz oranları ve kemoilüminens yoğunluğu saptandı.Sonuçlar: KIS grubu, normal kontrol grubu ile karşılaştırıldığında yaş ile daha az kilokazanımı, sükroz alımında azalma ve açık-alan araştırmada isteksizlik gösterdiler. KISbasamak-geçiş testinde, öğrenme belleğinde bir defisiti yansıtan olumsuz bir performansetkisi gösterdi. KIS grubu solda aşırı hipokampal kemoilüminens yoğunluğu ve apopitozoranları yansıtırken sağda önemli bir değişiklik göstermedi.Yargı: Bu bulgular sıçanlarda KISin hipokampüste asimetrik hasarlar ile öğrenme vebellekte bir defisite neden olabileceğini gösterdi.

Abstract

Background: Major depressive disorder (MDD) is associated with deficits in memory whichis often caused by hippocampal damages. Left-right asymmetry of human brain has beenunderstood for a century. Chronic mild stress (CMS) protocol is widely used to evokedepressive-like behaviors in laboratory rats. Few studies have been done to examine therelation between depression and left-right asymmetry hippocampal damages. This paperaimed to investigate the impacts of CMS on both sides of the hippocampus, learning andmemory.Methods: Body weight and sucrose preference and a modified open-field test was performedto check the depressive-like behaviors of CMS model. Learning and memory, apoptosis ratesand chemiluminescence density of the hippocampus were determined.Results: Compared with normal control, CMS group showed less weight gain with age,decreased sucrose intake and reduced open-field exploration indicative of depression; CMShad a negative effect on performance in the step-through test, reflecting a deficit in learningmemory; CMS group had a greater increase in the apoptosis rates and chemiluminescencedensity in the left hippocampus, but with no significant change in the right one.Conclusions: These findings suggest that CMS may cause the asymmetrical damages in thehippocampus which induce a deficit in learning and memory in rats.
