Selegiline Protects Motoneurons at Acute Phase of Spinal Cord Injury in Hemisectioned rats

Özet
Selegilin serbest radikal karşıtı ve nöroprotektif faktörleri indükleyen nöroprotektif veantiaptotik özelliklere sahiptir. Sıçanlarda hemiseksiyone edilen spinal kord selegilinin spinalkord yaralanmasının erken fazındaki etkileri değerlendirilmek için kullanıldı. Doz yanıtlarınıçalışmak üzere yedi grup oluşturuldu. İntraperitoneal 2,5mg/kg dozu en iyi yanıtın alındığıdoz olarak seçildi. Nöroprotektif etki değerlendirmesi için üç grup kullanıldı; bir grup sham- operasyonlu (SO) iken diğer iki grup T13 spinal segmental seviyede yapılan hemiseksiyon ilespinal yaralanmaya maruz bırakıldılar. Bir grup (HT) 21 gün boyunca 2,5mg/Kg dozundaselegilin ile tedavi edilirken diğer grup (HU) kontrol olarak hiç tedavi almadı. Tüm gruplarBasso Beattie Bresnahan (BBB) davranışsal testi kullanılarak değerlendirildi. Spinal dokularişlendi seri olarak kesitler alındı ve Cresyl violet ile boyandı. HT ve HU grupları arasındatotal motornöron sayımında (TMC) belirgin farklılık vardı. İyileşmeyi değerlendirmek içinnöroproteksiyon indeksi ve iyileşme indeksi olarak iki belirteç kullanıldı ve bunların değerlerisırasıyla 2,72 ve 3,8 olarak bulundu. Şifa indeksi HT ve HU gruplarındaki BBB skorlarınınbilgileri Gompertz modeli kullanılarak hesaplandı.HT ve HU grupları için sırasıyla eEnyüksek BBB skoru 13,75 ve 9,33; maksimum yanıt 5,3 ve 5,16; ve iyileşme oranı katsayıları20,6 ve 5,32 oldu. Akut spinal kord yaralanmasında selegilinin nöroprotektif olduğu yargısınavarıldı.
Abstract
Selegiline has neuroprotective and antiapoptotic properties including anti-free radical andneurorophic factors induction. Hemisectioned spinal cord of rat was used to evaluate theeffect of selegiline on the acute phase of spinal cord injury. Seven groups were used forstudying the dose response, where 2.5 mg/kg (intraperitoneal) was selected as the best dose.Three groups were used for evaluating the neuroprotectivity, one group was sham-operated(SO) while the other two groups were subjected to spinal cord hemisection, which was doneat T13 spinal segment level. One group (HT) was treated daily with 2.5 mg/kg selegiline for21 days while the other was untreated group (HU). All groups were evaluated using BassoBeattie Bresnahan (BBB) behavioral test. Spinal tissues were processed, serially sectionedand stained with Cresyl violet. There were significant differences in the total motoneuroncount (TMC) between HT and HU. Two indices were used to evaluate the improvement: themorphometric neuroprotection index and the recovery index (2.72. and 3.8, respectively). Therecovery index was calculated using Gompertz model for the data of BBB scores in HT andHU, the maximum BBB scores were 13.75 and 9.33; the times of maximum response were5.3 and 5.16; and the recovery rate coefficients were 20.6 and 5.32 for HT and HU,respectively. The conclusion is that selegiline is neuroprotective to the acute spinal cordinjury.
