Immobilization Stress in Pregnancy and the Maternal Brain's Response: an Experimental Study

Özet
Amaç: Çeşitli stres faktörlerinin anne ve fetusa kalıcı ve önemli zararlar verdiğigösterilmiştir. Bu çalışmada da kronik hareketsizlik stresi uygulanmış gebe farelerinbeyinlerinin hücre ölümü mekanizması açısından histolojik olarak incelemesi planlandı.Yöntem: Çalışmamızda 20-25 g arası 21 adet dişi ve katım için 25-30 g arası 5 adet erkekBalb/c fare kullanıldı. Çalışmamızda üç grup oluşturuldu. Grup 1: Kontrol Grubu (n=7), Grup2: Gebe grubu (n=7) Grup 3: Gebe Kronik Stres Grubu (n=7). Grup 1'deki deneklere hiçbirşeyuygulanmadı ve normal beyin histolojisi değerlendirildi. Süperovulasyon sonrası kronik stresuygulanan grupta ise dişi fareler gebeliklerinin 6. ve 18. günleri arasında günde 3 defa olmaküzere, her gün aynı saatte (09:00/12:00/17:00) 45'er dakikalık kronik hareketsizlik stresiuygulandı.Sonuç: Annelerden alınan beyin dokuları rutin histolojik parafin takibinden geçirildi vekesitler alındı. Prefrontal kortex ve hipocampus (CA1, CA2, CA3, GD) örneklerinde H&E,Cresly violet boyası sonrası genel inceleme ve nöron sayımı yapıldı. Apoptotik hücredeğişimlerini değerlendirmek amacıyla da aktive caspase-3 immunohistokimyası ve TUNELboyaması yapıldı.Tartışma: Çalışmamız anneye uygulanan fiziksel stresin beynin prefrontal korteksinde vehipokampal bölgelerinde belirgin nöronal azalmaya ve önemli miktarda apoptotic hücreartışına neden olduğu gösterilmiştir.
Abstract
Objective: Various stress factors have been shown to cause significant and permanent injuryto both mother and foetus. The histological examination of the brains of pregnant miceexposed to chronic immobilization stress in terms of the mechanisms of cell death was thepremise of this study.Methods: Twenty-one female mice weighing 20-25 g were used in the study, and 5 maleBalb/c mice weighing 25-30 g were used for mating. The study mice were divided into threegroups: Group I: Control Group (n=7), Group II: Pregnant Group (n=7), and Group III:Pregnant Chronic Stress Group (n=7). No intervention was performed on the mice in Group 1and normal brain histology was assessed. Female mice assigned to the chronic stress groupwere exposed to 45 minutes of chronic immobilization stress every day between gestationaldays 6 and 18, at the same hours three times a day (09:00,12:00 and17:00).Results: The brain cells obtained from the mothers were analyzed using routine histologicalparaffin and slices were obtained. The samples from the prefrontal cortex and thehippocampus (CA1, CA2, CA3, GD) were analyzed upon H&E and Cresly violet staining,and general analysis and neuron counts were determined. In addition, activated caspase-3immunohistochemistry and TUNEL staining were implemented in order to evaluate apoptoticcell changes.Conclusions: Our study demonstrated that maternal physical stress caused a significantincrease in the number of apoptotic cells along with a significant decrease in neuronal countin the prefrontal cortex and hippocampal regions of the brain.
