PI3K/Akt Inhibition Suppressesexcitatory Synaptic Transmission in Substantia Gelatinosa Neurons in an Inflammatory Pain Model in Rats

Özet

PI3K/Akt sinyal yolağının ısı ve mekanik hiperlagide rol aldığı gösterilmiştir. Nosiseptifeksitatör sinaptik geçiş, ağrı davranış yanıtlarında kritik bir faktördür. Bununla birliktePI3K/Akt yolağının spinal kord içinde eksitatör sinaptik geçişin modülasyonunda yer alıpalmadığı konusu çok net değildir. Bu çalışmada PI3K/Akt yolağının iki inhibitörü olanLy294002 ve wortmanninin eksitatör sinaptik geçişteki olası işlevsel rolleri araştırıldı.Ly294002 ve wortmannin tuzlu su enjeksiyonundan sonra erişkin sıçan spinal kordkesitlerinde spontan eksitatör postsinaptik akım (sEPSC) genliğinin frekansında birdeğişikliğe neden olmadı ancak herikisi de tam Freund adjuvan (CFA) enjeksiyonundan sonraerişkin sıçan spinal kord kesitlerinde sEPSC genliğinin frekansında azalmaya neden oldu.Dahası bu çalışma Ly294002 ya da wortmann tedavisi spinal kord dorsal kolonunda CFA ileoluşturulan artmış p-Aktin ekspresyonunu hafifletti. Böylelikle bu çalışma hipersensitifdurumda nosiseptifgeçişin sürdürülmesinde PI3K/Akt yolağının önemli bir rol oynadığınıgöstererek davranışsal noniseptif yanıtlara katkıda bulundu.

Abstract

PI3K/Aktsignaling pathway has been shown to contribute to thermal and mechanicalhyperalgesia. Nociceptive excitatory synaptic transmission is a critical factor to pain behaviorresponses. However, it is unclear whether PI3K/Akt pathway is involved in the modulation ofexcitatory synaptic transmission within the spinal cord. In this study, two inhibitors of thePI3K/Akt pathway, Ly294002 and wortmannin, were used to test the potential functionalroles in the excitatory synaptic transmission. Ly294002 and wortmannin did not change thefrequency or amplitude of spontaneous excitatory postsynaptic current (sEPSC) in the spinalslice preparedfrom adult rats by injection of saline, but bothof them decreased the frequencyand amplitude of sEPSC in the spinal slice preparedfrom adult rats by injection of a completeFreund's adjuvant (CFA). Further studyshowed thatLy294002 or wortmannin treatmentattenuatedCFA-inducedincreased expression of p-Aktin the spinal dorsal horn. Thus, thepresent study showed that PI3K/Akt pathway played an important role in themaintenanceofthe nociceptive transmission in hypersensitive state, contributing to behavioral nociceptiveresponses.
