Reactive Oxygen Species Mediate Abnormal Tau Phosphorylation in a Zinc-induced Model

Özet

Alzheimer Hastalığı (AD) olgularında amiloid-beta, reaktif oksijen gruplarının (ROS) aşırıoluşumuna neden olarak hipokampal nöronları saran oksidatif stres aşırı yüklenmesine yolaçtığı iyi bilinmektedir. Bir önceki çalışmamız insan nöroblastoma SH-SY5Y hücrelerindeluteolin (Lu)nin ROS-a bağlı bir tarzda çinko-bazlı tau fosforilasyonunu inhibe ettiğinigösterdi. Bu çalışmamızda ise daha ileri giderek bu modelde ROSun Tau fosforilasyonuüzerindeki etkilerini araştırdık. Ekzogenöz etkisi azaltılmış glutasyon ile hücrelerininkübasyonunun Ser262/356da tau fosfolilasyonun artışını iptal etmesiyle sonuçlandığınıgözlemledik. Dahası Lu veya Tau hiperfosforilasyonun inhibe edici ve altında kesinlikle yatanmekanizmayı açıklayıcı aksiyonu üzerine ROSun etkilerini araştırdık. Bulgularımız (1) GSHveya Lu ile ROS aşırı yapımının inhibisyonu Ser262/356 epitomunda tau fosforilasyonunudüzeltir, sırayla hidrojen peroksitin ortama eklenmesiyle hücre içi ROS artışı taufosforilasyonu üzerine Lunun inhibitör etkisini tersine çevirir; (2) hidrojen peroksit etkili birşekilde protein fosfataz 2Anın yapımını değiştirir.

Abstract

It has been well-established that amyloid-beta could induce reactive oxygen species (ROS)overproduction and lead to oxidative stress whose end-productions surround hippocampalneurons in Alzheimer's disease (AD) patients. In our previous study, it demonstrated thatluteolin (Lu) inhibited zinc-induced tau phosphorylation in an ROS-dependent manner inhuman neuroblastoma SH-SY5Y cells. In the current study, we further examined the effect ofROS on tau phosphorylation and its possible underlying mechanism in this model. We foundthat incubation of cells with exogenous reduced glutathione resulted in abolishment of theelevation of tau phosphorylation at Ser262/356. Moreover, we have further investigated theeffect of ROS upon the inhibitory action of Lu on tau hyperphosphorylation and it's a certainunderlying mechanism. Our data suggest (i) that inhibition of ROS overproduction by GSH orLu recovers abnormal tau phosphorylation at Ser262/356 epitope, in turn, the increase ofintracellular ROS by exogenous addition of H2O2 reverses the inhibitory effect of Lu on tauphosphorylation; (ii) that H2O2 efficiently changes the expression of the protein phosphates2A.
