Protective Effects of Vitamin D3 on Anxiety-like Behavior and the Total Antioxidant Power Following the Local Injection of Lysophosphatidylcholine in the Adult Rat Dorsal Hippocampus

Özet

Giriş: Çalışmalar multiple sklerozis (MS) hastalarında çeşitli düzeylerde anksiyetebulunduğunu ortaya koymuştur. Anksiyete üzerinde hippokampusun da etkisi vardır. Buçalışmada lysophosphatidylcholine (LPC) ile demyelinizasyon indüklenen sıçanlarda D3vitamininin anksiyete benzeri davranışlar üzerine olan etkisi ve dorsal hippokampuste (CA1alanı) total anti-oksidan gücü araştırılmıştır.Yöntem ve olgular: Hayvanlar dört gruba ayrılmışlardır. Her grupta 8 hayvan vardı. Kontrolgrup: CA1 alanına 2 μl saline verilmistir; sham grubu: intra-peritoneal yolla 150 μl susamyağı verilmistir; LPC grubu: stereotaktik yolla intra-hippokampal olarak CA1 alanına 2 μlLPC verilmistir; D3 vitamini grubu: LPC verildikten 7, 14 ve 21. Gün sonra 5 μl/kg dozda intra-peritoneal olarak D3 vitamini uygulanmıştır. Anksiyeteyi ölçmek üzere yükseltilmişlabirent (EPM) kullanılmıştır. Serbest radikallere karsı D3 vitamininin etkisinideğerlendirmek üzere FRAP tahlili uygulanmıştır.Bulgular: Bilateral LPC enjeksiyonu demyelinizasyona neden olmuştur. D3 vitaminiverilmesi %OAT ve %OAE oranlarını arttırmıştır. D3 vitamini verilmesi ayrıca total anti- oksidan gücü arttırmıştır.Sonuç: D3 vitamini uygulanması total anti-oksidan güç üzerine olan pozitif etkisi dolayısıylaacık anksiyolitik etkilere neden olabilmektedir.

Abstract

Background: Studies have reported levels of anxiety in multiple sclerosis (MS) patients.Considering the involvement of hippocampus in anxiety, an attempt is made in this study toinvestigate the effects of vitamin D3 on anxiety-related behavior and total antioxidant powerin the dorsal hippocampus (CA1 area) following the induction of demyelination bylysophosphatidylcholine (LPC).Methods: Animals were divided into four groups; each group contained eight animals.Control group: animals received 2 μl saline in CA1 area; sham group: animals received 150 μlsesame oil intraperitoneally; LPC group: animals received LPC by stereotaxic intra- hippocampal injection of 2μl LPC in CA1 area; Vitamin D3- treated group: animals weretreated with vitamin D3 at doses of 5μg/kg intraperitoneally for 7, 14 and 21 days afterreceiving LPC. Elevated plus maze (EPM) was used for testing the anxiety. The ferricreducing ability of plasma (FRAP) assay is chosen to assess the free radical scavenging powerof vitamin D3.Results: Bilateral injection of LPC caused demyelination and induced an anxiogenic-likeeffect which led to a reduction in the open arm time percentage (OAT%) and open arm entriespercentage (OAE%). Vitamin D3 administration increased the OAT% and OAE% withouteffects on locomotor activity compared to LPC-treated rats. Also, our results indicate that thevitamin D3 administration caused an increase in the total antioxidant power.Conclusion: It can be concluded that the vitamin D3 administration can produce clearanxiolytic effects through its positive effects on the total antioxidant power.
