Decrease of DISC1 Gene Expression is Associated With Increase of GSK3Beta Expression and Memory Loss of Mice Upon Aging

Özet

Giriş: Yaşlılarda yaşa-bağlı bellek yitimi oldukça yaygındır. Yaşlı nüfusun giderek artmasısonucu bu sorun giderek daha da ağırlaşmaktadır. Altında yatan moleküler mekanizmalar tamolarak anlaşılamadığı için etkili bir tedavi de şuan için olası değildir. Bu çalışma yaşlıfarelerde bellek bozukluğunun altında yatan mekanizmaları araştırmayı amaçlamaktadır.Yöntemler: Yaşlı sıçanlarda belleği incelemek üzere Y-maze spontan değişim paradigm veolfaktor bellek testleri kullanıldı. Hipokampüste DISC1 ve GSK3β varlığını göstermek üzereRT-PCR ve western blot teknikleri uygulandı. Kültüre edilmiş hipokampüste ve hipokampüsnöronlarında DISC1in fazlalığı ve sırasıyla siRNA teknikleri ve viral transfeksiyonlayokedidi.Sonuçlar: Yaşlı sıçanlarda bellek büyük oranda bozuktu. DISC1 önemli ölçüde azaldı, bununyanı sıra gençlere göre yaşlı fare hipokampüsünde GSK3β varlığı dramatik olarak arttı. Yaşlıfarelerde bellek bozuklğu DISC1 miktarının azalması ile ve GSK3β miktarının ise önemlimiktarda artması ile orantılı idi. Dahası kültüre edilmiş hipokampüsteki nöronlardaki DISC1in fazlalığı GSK3β azalmasına neden oldu. Nöronal ve glial hücre serilerinde DISC1yokluğu GSK3β artışına neden oldu.Yargı: Hepsi birden düşünüldüğünde bu sonçların, DISC1 varlığının ve aynı tarzda GSKβekspresyonunun yaşlılıkta bellek yitimi ile ilişkili ve payı olduğunu gösterir.

Abstract

Background: Age-related memory loss is quite common in the elderly. With the increase ofaged population, this problem is becoming more and more severe. The effective treatment iscurrently unavailable because of the lack of understanding of the underlying molecularmechanisms. The present study aimed to explore the mechanism underlying memoryimpairment in aged mice.Methods: Y-maze spontaneous alternation paradigm and olfactory memory test wereemployed to examine the memory in aged mice. RT-PCR and western blot were used todetect the expression of DISC1 and GSK3β expression in the hippocampus and culturedhippocampal neurons, in which DISC1 was overexpressed and knocked down using viraltransfection and siRNA techniques, respectively.Results: The memory was greatly impaired in aged mice. DISC1 expression was greatlydecreased, whereas the expression of GSK3β was dramatically increased in the hippocampusof old mice as compared to young mice. The memory impairment in old mice is highlycorrelated with decreased expression of DISC1 and increased expression of GSK3β.Furthermore, over expression of DISC1 in cultured hippocampal neurons resulted indecreased GSK3β expression, whereas knockdown of DISC1 increased GSK3β expression inneuronal and glial cell lines.Conclusion: Taken together, these results suggest that decrease of DISC1 expression and thesubsequent increase of GSK3β expression may contribute to the memory loss upon aging.
