Konteynerde saklanan donmuş boğa spermasına çevre faktörlerinin etkileri

Özet

Çalışmada aynı bakım ve besleme koşullarında barındırılan 5 baş Holştayn ırkı boğaya ait, 0.25 ml'lik payetlerde dondurulmuş spermalar kullanıldı. Aynı marka ve özellikte olan ve her biri beşer adet kanişler taşıyan iki saha konteynerine payetler paylaştırıldı. Bir kanistere her boğadan 20'er adet olmak üzere 100 payet, bir konteynerde bulunan 5 kanistere 500 ve 2 konteynere toplam 1000 payet depolandı. Konteyner kapasitesine göre sıvı azot miktarlarının; l no'Iu konteynerde 4/5 ve iki no'Iu konteynerde yaklaşık 1/4 olması sağlandı. Her iki konteynerde bulunan kanisterlere aynı işlemler uygulandı, l no'lu kanisterlerdeki payetler, konteyner ağzı içinde 5 sn bekletildikten sonra alındı. 2 no'lu kanisterlerdeki payetler, konteyner ağzı içinde 20sn bekletildikten sonra alındı. 3 no'lu kanisterlerdeki payetler, konteyner ağzı dışına 8-10 cm kadar çıkarıldı ve 5 sn bekletildikten sonra alındı. 4 no'lu kanisterlerdëki payetler konteyner ağzı dışına 8-10 cm kadar çıkarıldı ve 20 sn bekletildikten sonra alındı. 5 no'lu kanisterler kontrol grubu olarak bırakıldı ve içindeki payetlere çalışma sonuna kadar her hangi bir uygulama yapılmadı. İlk dört kanisterdeki payetler incelendikten sonra kontrol grubundaki payetler, kanister konteyner ağzından dışarı çıkarılmadan hemen alındı. Değişik kanisterlerden belirtilen uygulamalar sonrası alınan payet içindeki donmuş spermalar 37°C'lik su banyosunda 30 sn süresince eritildikten sonra spermatolojik özellikleri incelendi. Sonuç olarak; kanisterin kullanılmasına bağlı olarak oluşan ısı farklılıklarının spermatozoon motilitesine zarar verdiği, oluşan bu ısı farklılıklarının morfolojik bütünlüğü etkilemediği, kanisterin kaldırıldığı yükseklik, geçen süre ve konteynerlerdeki farklı azot miktarlarının spsrmatolojik özellikleri değiştirmediği saptandı.

Abstract

Summary: Semen was collected from 5 Holstein bulls under equal management conditions, extended with Laiciphos 478 and frozen in 0.25 ml straws. 200 straws from one ejaculate of every bull, total 100 straws in total 5 canisters in a container total 500 and in two containers total 1000 straws were stored. The containers capacities were 8 liters each. The first container (Container no 1) contained 4/5 of its liquid nitrogen capacity and the second (Container no II ) contained 1/4 of its liquid nitrogen capacity. Straws of the first container were taken after a 5 seconds waiting period in the neck-region of the container. Straws from the second container were taken out after a 20 seconds waiting period in the neck-region of the container. Straws from the 3rd container were taken after a 5 seconds waiting period at 8-10 cm out of the container. Straws of the 5th group were considered as control and no application was done until the end of the study. After the examination of the straws from the first four canisters, straws from the 5th container were taken out immediately without taking the canister out of the neck-region of the container. Frozen 5 straws from these different canisters were thawed for 30 seconds in 37°C water-bath and semen characteristics (motility, morphology) were evaluated. In conclusion; changes in the temperature relating to the usage of canister had detrimental effects on spermatozoa motility, this decrease in the spermatozoa motility was not affected by time and the distance of the canister taken out from the container. Changes in the temperature of the straws left in the canister while taking out of other straws had not no effect on morphology and the amount of liquid nitrogen in the container also had no effect on spermatozoa morphology.
