Comparison of the CMV scan assay system with CMV ELISA and CMV IFA immunoassay systems for detection of cytomegalovirus antibodies

Özet

Sitomegalovirus antikorlarının çabuk saptanması için 75 serum örneği üç test sistemi ile çalışıldı. İki aşamalı latex partikül aglutinasyonu olan CMV Scan testi, CMV IgG ve IgM antikorlarının saptanmasında kullanıldı. CMV antikorlarının saptanmasında CMV Scan testi, Vironostika CMV ELİSA (Organon Teknika) ve Fluoroset Immun Floresan antikor testi (Ortho Diagnostics) ile karşılaştırıldı. Çalışılan' 75 serum örneğinden 68 (% 91)'i üç test sistemi ile de aynı sonucu verdi. Sonuçlar karşılaştırıldığında üç test içinde duyarlılık ve özgüllük % 100/% 100 bulundu. Vironostika CMV ELİSA ile kuşkulu pozitif sonuçlar gözlendi. CMV İFA ile beta 2 mikroglobulin interferansı düşünüldü. CMV Scan ile sonuçlar daha kolay değerlendirildi. Vironostika CMV ELİSA ve Fluoroset CMV İFA, CMV Scan lateks aglutinasyon testi ile karşılaştırıldığında, her iki teste göre çabuk, uygulanabilir ve sonucu kolay değerlendirilebilir bir test olarak bulundu. Bu çalışmalar CMV Scan lateks aglutinasyon testinin klinik kullanımda uygun olduğunu gösterdi. CMV Scan çabuk ve kolay uygulanabilir olup Sitomegaloviruse karşı immün yanıtın taranmasında da yararlı bir test olabileceği sonucuna varıldı.

Abstract

Seventy five serum samples were tested with three methods for rapid detection of cytomegalovirus (CMV) antibodies. The CMV Scan, a two step latex particle agglutination assay for CMV IgG and IgM antibodies was evaluated for its ability to detect anti CMV antibodies. It was compared with the following assay systems: Vironostika CMV ELISA from Organon Teknika and Ortho Diagnostic Fluoroset Immunofluorescent Antibody Assay for CMV antibodies. Of the 75 clinical serum samples tested, 68 (91 %) gave concordant results among the three comparative assays. Comparison of results revealed a sensitivity / specificity of 100 %/100 % for the three assays. Border - line positive results were observed much more fre¬quently with Vironostika CMV ELISA and beta 2 microglobulin interference with Fluoroset CMV IFA than CMV Scan, compromising easy interpretation of results. CMV Scan is superior to Vironostika CMV ELISA and Fluoroset CMV IFA on the bases of performance characteristics, simplicity of test performance, and ease of interpretation of results. These studies indicate that the CMV Scan latex agglutination test is suitable for routine clinical laboratories. The CMV Scan test is rapid, self-contained, and fully practical for staff and would be useful for screening of cytomegalovirus immune status.
