Özet

Bu çalışma, bruselloz tedavisinde etkinliği kanıtlanmış olan rifampisin ile henüz etkinliği konusunda yeterli araştırma yapılmamış olan azitromisinin etkinliklerinin karşılaştırılması amacıyla yapılmıştır. Her biri 25-30 gram ağırlığında 52 erkek Balb/c deney faresine Brucella melitensis M16 suşu intraperitoneal yolla injekte edildi. İnokülasyondan yedi gün sonra dört fare eter anestezisi altında kesildi ve dalak kültürleri yapıldı. Kültür sonucunda tüm farelerin infekte olduğu saptandı. Kalan 48 fare üç gruba ayrıldı. Birinci gruba hiç bir ilaç verilmedi. ikinci gruba rifampisin, üçüncü gruba azitromisin verildi. Her iki ilaç, fareler 50 mg/kg/gün alacak şekilde hesaplanarak içme sularına konuldu. Tedaviye 15 gün devam edildi. Tedavinin bitiminden üç gün sonra fareler kesildi ve dalak kültürü yapıldı. Dalak homojenatının 1 mL' sinde üreyen koloni sayısı (CFU/mL) hesaplandı. Abstract

This study was designed to compare the efficacy of rifampicin the effect of which against brucellosis is well known, with azithromycin, the efficacy of which against brucellosis is not studied thoroughly, in the treatment of brucellosis. A total of 52 male Balb/c mice with average body weight of 25-30 g were used for this study, Brucella melitensis M16 strain was injected to the animals intraperitoneally. Seven days after the inoculation, four mice were decapitated under ether anaesthesia and spleen cultures were carried out. Culture results confirmed that all the animals were infected. The remaining 48 mice were divided into three groups. No treatment was given to the first group. Rifampicin and azithromycin were administered to the second and third groups, respectively. Both drugs were added to the drinking water as 50 mg/kg daily doses for 15 days. Following three days of the treatment, mice were decapitated and spleen cultures were carried out. Number of colony forming units grown in culture per mL (CFU/mL) of spleen homogenate was counted. It was seen that all animals in the control group were infected. Average LogW CFU/mL of this group was 4.51. There was no growth in both azithromycin and rifampicin groups. This difference was significant when compared with the control group (p
