Özet

Tüberküloz basilinin immünolojik özellikleri hücre duvarı ile ilişkilidir. Saflaştırılmış Mycobacterium tuberculosis (MTB) hücre duvarının moleküler yapısını aydınlatabilmek için pek çok teknik geliştirilmiştir. Bu şekilde analizler, basilin hücre duvarı ürünlerinin antijenitesinin, immünojenite, patojenite, bakteriyel fizyolojisinin ve diğer bazı bakteriler ile karşılaştırılmasını mümkün kılmıştır. Tüberküloza yönelik serolojik tanı testleri ve aşı geliştirmek için basilin protein antijenlerinin çalışılması önemlidir. Öte yandan, mikobakterinin moleküler yapısının kompleks olması, kültür nitratlarından ve hücresel ekstraksiyonlardan saflaştırılmasını güçleştirmektedir. Bu çalışmanın amacı, MTB'in protein antijenlerinin kromatografik teknik ile incelenmesidir. Mycobacterial antijenlerin geniş çeşitlilikte irdelenmesi amaçlandığı için üç ayrı (BCG, PPD, sonikat) antijen araştırılmıştır. Bu amaçla bir seri her üç antijene yönelik yüksek performans likid kromatografi (HPLC) ve enzime bağlı irnmun test (ELISA) çalışılmıştır. Sonuç olarak, başlıca insanda ısı şok proteini eşdeğeri olan 65kD her kromatogramda gözlenmiştir. Ayrıca Türkiye'ye farklı zamanlarda giren BCG aşılarından farklı protein pikleri elde edilmiştir. Bunların açıklaması daha ileri çalışmaları gerektirmektedir.

Abstract

Important protein-based immunoreactivities have long been associated with the cell wall core of mycobacteria. In order to explore the molecular basis of such activities, purified cell walls of Mycobacterium tuberculosis (MTB) were extracted with many techniques. After these kinds of evaluation, the antigenity of these products can be demonstrated and also their roles in immunogenicity, pathogenicity, and bacterial physiology and comparison to analogous polypeptides from other bacteria can be discussed. To develope serodiagnosis or vaccine, protein analysis is essential. The complex nature and the chemical diversity which characterize the antigens of pathogenic mycobacteria have, however, limited the purification of it from culture filtrates and cellular extracts. The aim of this study is to evaluate the protein antigens of MTB by chromatographic techniques. To cover a spectrum of different mycobacterial antigens, BCG, PPD, sonicate antigens were subjected to series of High Performance Liquid Chromatography (HPLC) and the fractions obtained were used to detect serum antibodies using Enzyme Linked Immun Assay (ELISA). In conclusion, mycobacterial 65kD which is homologous to human 60kD heat schock protein is predominant in all three MTB antigens. Different protein peaks have been shown in different kinds of BCG vaccines which need further evaluation.
