Comparison of different methods for the detection of beta-lactamase activity in Candida albicans strains

Özet

Bu çalışmanın amacı, Candida albicans'ta beta-laktamaz saptamak için kullanılan iki yöntemi karşılaştırmak idi. Çeşitli klinik örneklerden soyutlanan 154 C. albicans ve bir ATCC (C.albicans ATCC 90028) suşunda beta-laktamaz üretimi nitrosefin ve iyodometrik yöntemler kullanılarak araştırıldı. Nitrosefin yöntemiyle ilk üç saatte suşların dördünde (%2.6), 24 saatte ise suşların beşinde (%3.2) beta-laktamaz varlığı saptandı. İyodometrik yöntem uygulanıp renk değişikliğinin gözle ve spektrofotometrik değerlendirilmesiyle izolatların sırasıyla beşi (%3.2) ve 20'si (%13.0) olumlu bulundu. İyodometrik yöntemin gözle değerlendirilme sonuçları, referans yöntem olarak nitrosefin yönteminin 3. saat sonuçlarıyla karşılaştırıldığında duyarlılık, özgüllük, pozitif ve negatif prediktif değerler sırasıyla %100, %99.3, %80, %100 olarak saptandı. Aynı bulgular, nitrosefin yönteminin 24. saat sonuçlarıyla karşılaştırıldığında ise tüm değerler % 100 olarak bulundu. Bu değerler, spektrofotometrik değerlendirmenin nitrosefin yöntemiyle karşılaştırılmasında ise sırasıyla %20, %87.2, %5, %97 olarak bulundu. Bu sonuçlar ışığında iyodometrik yöntem uygulanıp renk değişikliğinin gözle değerlendirilmesinin C. albicans suşlarında beta-laktamaz varlığının saptanmasında, güvenilir, hızlı ve ucuz bir yöntem olduğu sonucuna varıldı.

Abstract

The purpose of the present study was to compare two methods for detection of beta-lactamase in Candida albicans. Beta-lactamase production was searched in 154 C. albicans strains isolated from various clinical specimens and one ATCC strain (C. albicans ATCC 90028) by using nitrocefin and iodometric methods. Four of the strains (2.6%) revealed the existence of beta-lactamase within three hours while five of them (3.2%) were positive after 24 hours by nitrocefin method. When the visual and spectrophotometric values for the iodometric method were considered, beta-lactamase activity was detected in five (3.2%) and 20 (13.0%) of the strains, respectively. The sensitivity, specificity, positive predictive and negative predictive values were found to be 100.0, 99.3, 80.0, 100.0%, respectively by visual evaluation of the iodometric method when compared to nitrocefin test after three hours as the reference method. They were all 100.0% when compared to nitrocefin test after 24 hours. These values were calculated to be 20.0, 87.2, 5.0, 97.0%, respectively, for the spectrophotometric evaluation. As a result, it can be concluded that the iodometric method and visual interpretation is a reliable, rapid and cheap technique for the detection of beta-lactamase production.
