Non-conjugative plasmids encoding TEM-type beta-lactamase in clinical isolates of Escherichia coli

Özet
Bu çalışmanın amacı, Escherichia coli'de TEM-tipi $beta$-laktamaz kodlayan non-konjugatif plazmitleri araştırmak idi. Altmış ampisilin dirençli E. coli kökeni TEM-tipi $beta$-laktamaz kodlayan plazmitler açısından tarandı. Konjugasyon deneyleri ile altı susta >48 kbp molekuler ağırlıklı konjugatif plazmitler belirlendi. Escherichia coli K-12 suşu J53-2 transkonjugantlarının hepsinde çift disk sinerji testi ile genişlemiş-spektrumlu $beta$-laktamaz (GSBL) üretimi belirlendi; GSBL üretmeyen geri kalan 54 E. coli'den hazırlanan plazmit DNA preparatları transformasyon deneylerinde kullanıldı. Elde edilen dört transformanttan ayrı ayrı yaklaşık olarak 3, 3.8, 13 ve 18 kbp'lik dört non-konjugatif plazmit izole edildi. Polimeraz zincir reaksiyonunda (PZR) altı konjugatif ve dört non-konjugatif plazmitte TEM-tipi gen pozitifti. Transformantların standart disk difüzyon testi ve PZR analiz sonuçlarına göre, dört non-konjugatif plazmidin TEM-1 veya TEM-2 tipi geniş spektrumlu $beta$-laktamaz enzimi kodladığı düşünüldü. Geniş spektrumlu $beta$-laktamaz üreten transformantların birinin TEM-tipi $beta$ -laktamazları aşırı miktarda ürettiği düşünüldü. Ampisiline dirençli E. coli suşlarının molekuler epidemiyolojisinde konjugatif plazmitler yanı sıra non-konjugatif plazmitlerin de tiplendirmesi yararlı olacaktır.
Abstract
Sixty ampicillin-resistant Escherichia coli isolates were screened for the plasmids encoding TEM-type of $beta$-lactamase. In six strains, conjugative plasmids of about >48 kbp in molecular weight were detected by conjugation experiments. All of the E. coli K-12 strain J53-2 transconjugants were determined to produce extended-spectrum $beta$-lactamases (ESBL) by double disk synergy test. Plasmid DNA preparations from the remaining 54 E. coli strains with no ESBL were used in transformation experiments. Four non-conjugative plasmids of about 3, 3.8, 13 and 18 kbp were separately isolated from four transformants obtained. Six conjugative and four non-conjugative plasmids were positive for TEM-type $beta$-lactamase genes in polymerase chain reaction (PCR). According to the results of disk diffusion test and PCR analysis of transformants, four non-conjugative plasmids were thought to encode TEM-1 or TEM-2 type of broad-spectrum $beta$-lactamases. One of the broad-spectrum $beta$-lactamase producing transformants was thought to be a hyperproducer of TEM-type of beta-lactamase. It is concluded that typing of non-conjugative plasmids other than conjugative plasmids could also be useful in molecular epidemiology of ampicillin resistant E. coli strains.
