Özet

Brusellozun klinik evresini tanımlamada serolojik yöntemler önemlidir. Klinik olarak bruselloz öntanısı konulmuş hasta serumlarıyla yapılan bu çalışmada, rivanol testi ve ELISA ile elde edilen sonuçların karşılaştırılması amaçlanmıştır. Çalışma, Rose Bengal (Chimica Diagnostic!, italya) pozitif 40 serum örneği ve kontrol grubu olarak sağlıklı 20 kişiden alınan serum örnekleri ile gerçekleştirilmiştir. Örneklere önce standart tüp aglutinasyon yöntemi (Bacterial Suspensions Tube Test/Biomedical Systems, ispanya) uygulanmış, pozitif olanlarda rivartoilü tüp aglutinasyon ve ELISA (Virotech/Almanya) ile IgM ve IgG taraması yapılmış; brusellozun tanısında iki testin etkinliği, birbiri ile uyumu ve kullanılabilirliği değerlendirilmiştir. Standart tüp aglutinasyon test sonuçları esas alındığında, rivanol testi ile ELISA IgG sonuçları arasında istatistiksel olarak anlamlı bir fark olmadığı ve bu çalışmada her iki yöntemin duyarlılığının %89, özgüllüğünün %100 olduğu saptanmıştır. Sonuç olarak; her iki yöntemin tanıda birbirine fazlaca bir üstünlüğünün olmadığı, ancak standardize edilmiş iyi bir ELISA kitinin IgM ve IgG antikorlarının birlikte pozitif olduğu örneklerde rivanol testine üstünlük sağlayabileceği düşünülmektedir.

Abstract

Serologic methods are important for the determination of the clinical stage of brucellosis. In this study, the diagnostic efficacy of ELISA (Virotech/Germany) results were compared with those of rivanol test in patients diagnosed as brucellosis. Serum samples of 40 Rose Bengal (Chimica Diagnostici/ltaly) positive patients were studied and as control of 20 healthy subjects. Firstly, standard tube agglutination test (STA) (Bacterial Suspensions Tube Test/Biomedical Systems/Spain) was performed for each sample. If the test was positive, IgG and IgM were evaluated by rivanol tube agglutination and ELISA. On the basis of STA results, there was no significant difference as to results between the rivanol test and ELISA IgG. The sensitivities and specificities of both tests were 89% and 100%, respectively. No test was found to be superior than the other, however, a standardised ELISA test might provide better evaluation than rivanol test for sera that have both IgG and IgM.
