Direkt HDL-kolesterol tayin yöntemiyle çöktürmeli HDL-kolesterol tayin yönteminin karşılaştırılması

Özet
Amaç: Bu çalışma serumda direkt HDL-Kolesterol tayin yönteminin doğruluğunu ve tutarlılığını saptamak amacıyla yapılmıştır. Yöntem: Bu amaçla 50 olguda direkt ve çöktürmeli yöntemlerle serum HDL-Kolesterol düzeyleri ölçülmüştür. Direkt yöntemin gün içi tutarlılığını saptamak için 5 olguda HDL-Kolesterol düzeyleri aynı anda 20 kez çalışılmıştır. Günler arası tutarlılığı ölçmek için 2-8°C'de saklanan örneklerde 12 gün boyunca direkt yöntemle HDL-Koleste-rol düzeyleri ölçülmüştür. Bulgular: iki yöntem arasındaki korelasyon katsayısını r:0.9938, doğru denklemini y=0.985 x -1.26 mg/dl olarak bulduk. İki yöntemin sonuçları ileri derecede uyumluydu. Direkt yöntemin gün içi ortalama C.V.'i %0.836 bulundu. Direkt yöntem ile 7. güne kadar ortalama C.V. %5 bulundu. Günler arası korelasyon 7. güne kadar anlamlıydı. Sonuçlar: Bu bulgularımıza dayanarak serumda direkt HDL-Kolesterol tayin yönteminin rutin laboratuar çalışmalarında güvenilir şekilde kullanılabileceği sonucuna vardık.
Abstract
Purpose: This study was done to determine the accuracy and precision of direct measurament method of HDL-Cholesterol. Method: HDL-Cholesterol levels in serum was determined with direct method and with precipitation method in 50 patients. To measure the accuracy of the direct method winhin a day we worked HDL-Cholesterol levels in 5 sampels 20 times at the same time. In day to day work for the accuracy we worked with 10 samples which were all stored at the temperatura of 2-8 C during 12 days. Results: In both methods correlations coefficient (r) was: 0,9938 and lineer regression was y= 0,985x -1,26 mg/dl. As a result a significant correlations was found between two methods. For direct method within a day mean value of C.V. was %0.836. For direct method we es-tablished the mean C.V. %5 after 7 days. Conclusions: According to the data given above we are cqnfident to recommend that direct measurament method of HDL-Cholesterol is suitable for rutin laboratories use.
