Kıymalarda E.coli 0157:H7 aranmasında EZ coli kiti kullanımı üzerine araştırma

Özet

Bu çalışmada 57 çiğ kıyma örneğinde standart kültürel yöntem ve EZ Coli hazır kiti ile E coliO157:H7 serotipi aranmıştır. EZ Coli kiti ile 2 adet örnekte (%3,5) pozitif sonuç alınmış olmasına rağmen pozitif sonuçların elde edildiği zenginleştirme besiyerinden SMAC ağar besiyerine yapılan ekimlerde E coliO157 serotiplerine rastlanılmamıştır. Benzer şekilde pozitif sonuç alınan örneğe ait 1ST broth ve mEC broth kültürlerinden de SMAC ağarda bu serotiplere rastlanılmamıştır. Bu bulgular, refakatçi flora baskılaması nedeni ile izolasyon yapılamaz iken kitin çok duyarlı oluduğunu göstermekle beraber, tersine olarak, bir başka örneğe ait EZ Coli zenginleştirme besiyerinden SMAC ağara yapılan ekimde E. coliO157 serotipinin izole edilebilmesi, buna karşın bu örnekte kitin negatif sonuç vermesi bu kez de kitin duyarılğı hakkında endişe doğurmuştur. Bu kit çok duyarlı olsa bile, pozitif sonuç alınan örneklerden standart kültürel yöntemlerle izolasyon yapılıp bakterinin H7 serotipi olup olmadığı ve/veya verotoksijenik olup olmadığı saptanamaz ise rutin kullanımda fazlaca bir şansı olmayacaktır.

Abstract

In this research, 57 ground beef samples were analyzed by EZ Coli kit and simultaneously by conventional culture techniques for the presence of E. coli 0157: H7 serotype. Although 2 positive results (3.5%) were obtained by EZ Coli kit, no E. coli 0157 isolation was succeeded from the SMAC agar dishes spreed from EZ Coli enrichment broth and also LST broth and mEC broth cultures of the same samples. These results indicate that this kit is very sensitive even the masking of cohabitant flora in agar media. On the contrary, one E. coli 0157 was isolated from EZ Coli kit enrichment broth culture while the kit gave negative result. Hence a suspicion was borne about the sensitiveness of the kit. Even if the kit is highly sensitive, if no isolation can be done by conventional techniques or it can not be determined whether the isolates are H7 serotype and/or verocytotoxigenic, the kit will have not have any chance to be employed for routine analysis.
