Özet

Amaç: Transfusion-transmitted virus (TTV) ilk olarak 1997 yılında, kan transfüzyonunu takiben non-A-G hepatitli hastalarda belirlenmiştir. Bu çalışmanın amacı TTV'nin ORF2b gen kısımlarının genetik klonlanmasıdır. Gereç ve Yöntem: Bu çalışmada TTV DNA pozitif hasta örneklerinden ORF2b gen kısmı polimeraz zincir reaksiyonu (PCR) ile çoğaltıldı ve pGEM®-T plasmid vektörde klonlandı. Bulgular: ORF2b genleri bulunduran rekombinant vektörler restriksiyon enzim analizi ve PCRtarama testleri ile belirlendi. Sonuç: TTV'nin genlerin klonlanması ülkemizde viral gen kütüphanelerinin zenginleşmesini sağlayacaktır. Ayrıca, gelecek çalışmalarda bu rekombinant plasmidlerle açıklatılacak hedef proteinler ELISA kitlerinin geliştirilmesinde kullanıla bilinecektir.

Abstract

Objective: Transfusion-transmitted virus (TTV) was first identified in patients with non-A to -G hepatitis following blood transfusion in 1997. The objective of this study is cloning of ORF2b gene fragments of TTV. Materials and Methods: In this study, the ORF2b genes from TTV DNA positive samples were amplified by polymerase chain reaction (PCR) and cloned into pGEM®-T plasmid vector. Results: The recombinant plasmid vectors including ORF2b gene were detected by PCRscreening test and restriction enzyme analysis. Conclusion: The producing of the gene clones containing ORF2b gene of TTV will contribute the enrichment of the viral gene library of our country. Moreover, the target proteins produced from the recombinant clones in future studies will be used in terms of the development of ELISA kits.
