Damızlık tavuk işletmelerinde tespit edilen mikoplazma infeksiyonları

Özet
Kanatlı hayvanlarda hastalığa neden olan farklı mikoplazma etkenleri bulunmaktadır. Bu etkenlerden Mycoplasma gallisepticum (M. gallisepticum) ve Mycoplasma synoviae (M. synoviae) en önemli türler olup OIE listesinde yer almaktadır. M. gallisepticum ve M. synoviae vertikal bulaşması nedeniyle damızlık sürülerde ayrı bir öneme sahiptir. Özellikle de vertikal bulaşmanın şekillenmesi ve damızlıkların bu infeksiyonlar yönünden ari olması gerektiği için, bu iki infeksiyon etkeninin hızlı, güvenilir bir testle teşhisi neticesinde acil koruma ve kontrol önlemlerinin alınması şarttır. Bu çalışmada 2009 yılında Merkez Veteriner Kontrol ve Araştırma Enstitüsü Kanatlı Hastalıkları Teşhis Laboratuvarı tarafından denetimi yapılan damızlık işletmelerinde polimeraz zincir reaksiyonu (PZR) ve etken izolasyonu ile tespit edilen mikoplazma infeksiyonları ortaya konmuştur. Enstitümüze bağlı illerde bulunan damızlık işletmelerden alınan kan serumu örnekleri M. gallisepticum ve M. synoviae yönünden serum pleyt aglutinasyon test (SPAT) ve ELISA ile test edildikten sonra, seroloji ile M. synoviae pozitif bulunan damızlık işletmelerden alınan trakeal svap örnekleri PZR ve etken izolasyonu ve identifikasyonu yapılarak incelenmiştir. Damızlık işletmelerin tamamı M. gallisepticum yönünden negatif bulunmuştur. İşletmelerin %8.5’i SPAT, ELISA ve PZR ile M. synoviae pozitif bulunurken ancak %2.5’inden etken izolasyonu yapılabilmiştir. PZR ve etken izolasyonu karşılaştırıldığında ise; PZR ile pozitif bulunan işletmelerin sadece %33.4’ünde etken izolasyonu yapılabilmiştir. M. gallisepticum ve M. synoviae’nın teşhisinde PZR tamamen konvansiyonel kültürel yöntemlerin yerini alamamasına rağmen, bakteriyoloji ile desteklendiğinde erken ve kesin teşhis açısından damızlık sürülerde başta olmak üzere kanatlı endüstrisine faydalı olacaktır.
Abstract
Various Mycoplasma species which causes illness at poultry exist. Of the all Mycoplasma spp. Mycoplasma gallisepticum (M. gallisepticum) and Mycoplasma synoviae (M. synoviae) are the most important species and take place in the OIE list. Due to vertical transmission of M. gallisepticum and M. synoviae, they do have an importance at breeder flocks. Urgent protection and control measurements have to be applied according to the results of the diagnosis of rapid and reliable test due to the vertical transmission and Mycoplasma-free required breeder flocks. In this study, Mycoplasma infections diagnosed by polymerase chain reaction (PCR) and agent isolation at the breeder flocks which are controlled by Poultry Diagnosis Laboratory of Central Veterinary Control and Research Institute were introduced. After the serum samples which had been taken from the breeder flocks in the provinces under the responsibility of our institute were tested for M. gallisepticum and M. synoviae by SPAT, ELISA, tracheal swap samples taken from M. synoviae positive breeder flocks were examined by PCR, agent isolation and identification. All the breeder flocks were found to be M. gallisepticum negative by SPAT and ELISA. Of the holdings, while 8.5% were found to be M. synoviae positive by SPAT, ELISA and PCR, agent identification was achieved from 2.5%. Comparison of PCR and bacteriology, agent isolation was done 33.4% solely from the holdings which are found to be M. synoviae positive by PCR. Although PCR can not replace the conventional cultural methods totally for the diagnosis of Mycoplasma spp. it will be useful for poultry industry, especially, poultry breeders, if it is supported by conventional cultural methods in order to rapid and definite diagnosis of Mycoplasma spp.
