NiFe2O4 Nanokompozitinin Olası Toksik Etkisine Karşı Olueropein'in Koruyucu Rolü

Özet
Nikel demiroksit (NiFe2O4) nanokompozitinin fotoelektrik cihazlar ve sensörlerin yapımı ile modern sanayi alanından manyetik rezonans görüntüleme ve hedeflenmiş ilaç tedavisi gibi biyomedikal uygulamalara kadar hayatımızın pek çok önemli alanında kullanıldığı bilinmektedir. Bu çalışmada, metal oksit nanopartiküller arasında yer alan NiFe2O4 (30 nm) nanopartikülünün Drosophila melanogaster'in yabanıl soyu üzerinde olası toksik etkisine karşı zeytin yaprağından elde edilen Oleuropein'in koruyucu etkisinin olup olmadığı araştırılmaya çalışılmıştır. Bu amaçla, D. melanogaster larva ve ergin bireylerine farklı konsantrasyonlarda NiFe2O4 (25, 50, 75, 100, 200 ve 500 µg/mL) ve Oleuropein (100 µM) uygulanarak yaşama yüzdesi ve yavru birey sayısı deneyleri yapılmıştır. Kontrol ve uygulama gruplarına ait sonuçların istatistiksel analizleri için tek yönlü varyans analizi (ANOVA) ve Duncan testi uygulanmıştır. Çalışma sonucunda yaşama yüzdesi deneylerinden elde edilen verilere göre, larvalara uygulanan NiFe2O4 nanopartikülünün tüm konsantrasyonları kontrole göre yaşam yüzdesini azaltırken, Oleuropein uygulaması yaşama yüzdesini arttırmıştır. Bununla birlikte, etkisi araştırılmak üzere bir deney setinde yalnızca ergin dişi bireylere, diğer deney setinde ise yalnızca ergin erkek bireylere NiFe2O4 ve Oleuropein uygulamalarının yapıldığı yavru birey sayısı deneyleri sonucunda, NiFe2Onanokompozitinin konsantrasyon artışına bağlı olarak F1 neslinde toplam yavru döl sayısının azaldığı, Oleuropein uygulanan deney setlerine ait F1 neslinde ise bu sayının artarak kontrole yaklaştığı görülmüştür (p< 0.05). Yaşama yüzdesi ve yavru birey sayısı deneylerinde NiFe2O4 nanokompozitinin toksik etkisine karşı serbest radikal oluşumunu engelleme ve detoksifikasyon enzimlerini uyarma özelliği bulunan Oleuropein'in koruyucu bir rol oynadığı düşünülmektedir.
Abstract
Nickel iron oxide (NiFe2O4) nanocomposites are known to be used in many important areas of our lives from photoelectric devices and modern industrial area to magnetic resonance imaging and to biomedical applications like targeted drug therapy. In this study, whether Oleuropein, derived from olive leaves have preventive effect on Drosophila melanogaster wild type strains against the possible toxic effects one of metal oxide nanoparticles, NiFe2O4 (30 nm) were investigated. For this purpose, different concentrations of NiFe2O4 (25, 50, 75,100, 200 and 500 µg/mL) and Oleuropein (100 µM) were used to investigate the percentage of life span and the number of the offspring on D. melanogaster larvae and adults. For statistical analysis of the control and treatment groups' results One-way analysis of variance (ANOVA) and based on data obtained from the input data, all applied concentrations of nanoparticles NiFe2O4 reduce the percentage of living when compares the control group, and applied Oleuropein increased the survival rate of the larvae. However, to investigate the effect of the NiFe2O4 and Oleuropein on the number of the offspring, in a set of experiment only adult female individuals were subjected to NiFe2O4 and Oleuropein while in the other set of experiment only male individuals do. In parallel with the increased concentration of NiFe2O4 reduce the number of offspring of the F generation in total and in Oleuropein applied groups it increased the number of offspring belong to F1 generation approaching the control (p< 0.05). In the lifespan and number of offspring studies, against the toxic effect of NiFe2O4 nanocomposites, Oleuropein is thought to have a protective role by of blocking the formation of free radicals and inducing the detoxification enzymes.
