Köpeklerde deneysel akciğer aspergillozis' i ve itrakonazol ile sağaltımı

Özet

Bu çalışmada, köpeklerde Aspergillus fumigatus ile intratrakeal yolla oluşturulan deneysel enfeksiyonda; klinik, hematolojik, biyokimyasal, idrar, histopatolojik ve mikolojik bulguların belirlenmesi ve itrakonazol'ün sağaltım etkinliğinin değerlendirilmesi amaçlandı. Çalışmada, 16 sağlıklı erkek köpek, enfekte kontrol ve sağaltım grubu olmak üzere iki eşit gruba ayrıldı. Aspergillus fumigatus ihokulasyonundan önce köpeklere siklofosfamid verildi. Daha sonra intratrakeal yolla A. fumigatus inoküle edildi. Etkenin verilişinin 5. gününde her iki grupta lökositosiz belirlendi. Serum ALP aktivitesi yükselirken, total protein, albumin ve globulin konsantrasyonları düştü. Köpeklerin akciğerler ve burun eküvyonundan A. fumigatus izole edilirken histopatolojik olarak sadece akciğerlerde granulomatöz yangı ve etken hifaları tespit edildi. Etken verilmesini takiben 7. günde sağaltıma başlandı. Sağaltım grubundaki köpeklere 10 mg/kg vücut ağırlığı/gün dozda 30 gün süreyle oral Itrakonazol verildi. Deneme sonunda, sağaltım grubunda bir köpekte gözlenen deri lezyonları ve enfekte kontrol grubunda iki köpekte gözlenen seröz burun akıntısı dışında tüm klinik belirtilerin düzeldiği belirlendi. Deneme öncesindeki değerlere göre, enfekte kontrol grubunda serum albumin konsantrasyonunda ve ALT aktivitesinde düşme, sağaltım grubunda serum ALT aktivitesinde yükselme, MCHC değerinde düşme ve total protein,.albumin ve globulin konsantrasyonlarında düşmenin devam etmesi dışında her iki grupta hematolojik ve biyokimyasal parametrelerin normale yakın olduğu tespit edildi. Enfekte gruptaki 2 köpeğin burun eküvyonu ve akciğerlerinden etken izole edilirken her iki gruptaki köpeklerde akciğerlerin histopatolojik olarak normal olduğu belirlendi.

Abstract

The aim of this study was to determine the clinical, haematological, biochemical, urine, histopathological and my-cological findings and'to evaluate the efficiency of itraconazole treatment in experimentally infected dogs via intratracheal route with Aspergillus fumigatus. In study, 16 healthy male dogs were evenly divided into two groups as infected control and treated. Cyclophosphamide was injected to dogs before the inoculation of A. fumigatus. Then, A. fumigatus was inoculated via intratracheal route. On day 5 after the inoculation of the agent, leukocytosis were determined in both groups. Serum total protein, albumin and globulin concentrations were decreased while serum ALP activity was increased. Aspergillus fumigatus was isolated from nasal swaps and lungs of the dogs while the hyphae of the agent and granulomatous inflamation were observed only in lungs by histopathological examination. The treatment of animals was started on day 7 after the inoculation of the agent. Itraconazole, at 10 mg/kg body weight/day was administrated orally for 30 days. At the end of the experiment, all of the clinical signs in both groups were recovered except cutaneous lesions in one dog in the itraconazole treated group and serous nasal discharge in 2 dogs in the infected group. Compared to the beginning values of the experiment, haematological and biochemical parameters in both groups returned closely to normal values except the reductions in serum albumin concentrations and ALT activities in infected control group; elevation in serum ALT activity; decrease in MCHC; progressive reduction in total protein, albumin and globulin concentrations in itraconazole treated group. Significant histopathologic lesions in the lungs of the dogs were not seen in both groups while Aspergillus fumigatus was isolated from nasal swaps and lungs from only 2 dogs in the infected group.
