N Nitroso-diethylamine (DENA) uygulanan ratlarda melatonin hormonunun koruyucu etkileri

Özet
N nitroso-diethylamine (DENA) organizmada karaciğer kanseri oluşturan bir maddedir. Epifiz bezinden salgılanan melatonin hormonu ise onkostatik etkileri olan ve serbest radikallerin dönüşüm reaksiyon basamaklarında olumlu rol oynayan özelliklere sahiptir. Bu çalışmada, Grup 1 deki erkek sıçanlara melatonin (1g/kg) + DENA (200mg/kg), Grup 2 deki erkek sıçanlara DENA (200mg/kg) uygulandı. 10 hafta sonra kontrol ve deney gruplarındaki sıçanlardan kan örnekleri alındı. Deney gruplarındaki kan örneklerinde saptanan serum AST ve ALT, LDH ve GGT değerlerinin kontrole göre yükselme gösterdiği bulundu. Alk. Phos değerleri Grup 1 'de yükselirken Grup 2'de kontrole göre düşüş göstermiştir. Serum kolesterol ve total protein değerleri deney gruplarında düşme gösterirken trigliserit değerlerinin arttığı saptanmıştır. Sonuç olarak, melatonin hormonu uygulamasının DENA'nın ortaya çıkardığı saptanan serum enzim seviye yükselmelerini engellemediği söylenebilir.
Abstract
N nitroso-diethylamine (DENA) is a material which causes liver cancer in organisms. Melatonin hormone which is secreted from ephiphysis has oncostatic effects and characteristics played a positive role in the neutralising steps of free radicals. In this study, melatonin (1g/kg) + DENA (200mg/kg) was treated to the male rats in Group 1, and DENA (200mg/kg) was treated to the male rats in Group 2. 10 weeks later blood samples were taken from rats in the control and experiment goups. In the serum samples of experiment group, it was observed that AST and ALT, LDH and GGT values have increased value compared to control group. When serum Alk. Phos. values increased in Group 1, in Group 2 it showed decreasing compare to control group. When serum cholesterol and total protein values, (in Group 1 and 2) shows decreasing compare to control group the trigliseride values are increasing. As a result, it shows that, with the practice of the DENA, 1 mg/kg (body -weight) melatonin hormone do not block the increasing value of the serum enzyme.
