Deltametrin'in Allium cepa L.'daki Fizyolojik, Anatomik ve Genotoksik Etkilerinin Araştırılması

Özet
Bu çalışmanın amacı, Allium cepa L.’da deltametrinin toksik etkilerini araştırmaktır. Bu amaçla, kök uzunluğu,ağırlık kazanımı, çimlenme yüzdesi, mikronukleus (MN), kromozomal anormallik sıklığı ve anatomik değişimlerile malondialdehit (MDA) düzeyi gibi parametreler kullanılmıştır. Bulblar 1 kontrol ve 3 uygulama olarak 4gruba ayrılmıştır. Kontrol grubu bulblar çeşme suyu, uygulama grubu bulblar ise deltametrinin 5, 10 ve 20 mg L-1dozları ile 72 saat muamele edilmiştir. Sonuçlar, kontrol grubu ile karşılaştırıldığında, tüm uygulamagruplarında, kök uzunluğunu, ağırlık kazanımı ve çimlenme yüzdesinin azaldığını, MN ve kromozomalanormalliklerin sayısının ise arttığını göstermiştir. Yapışkan kromozom, köprü, fragment, c-mitoz ve kromatinineşit olmayan dağılımı şeklinde kromozomal anormallikler gözlenmiştir. Ayrıca, sonuçlar, deltametrininuygulanan tüm dozlarda yassılaşmış hücre çekirdeği, nekroz, iletim dokuda bazı maddelerin birikimi, kortekshücre duvarında kalınlaşma, belirgin olmayan iletim doku ve hücre deformasyonu vb. anatomik hasarlara nedenolduğunu da göstermiştir. Diğer yandan, deltametrine maruz kalan kök uçlarının MDA düzeylerinde önemli birdeğişim olduğu saptanmıştır. Deltametrin uygulamasının lipid peroksidasyonunu önemli ölçüde arttırdığı ve tümuygulama gruplarında MDA düzeylerinde bir artışa neden olduğu belirlenmiştir. Bu çalışmada elde edilenveriler, A. cepa biyo-analizlerinin deltametrin’in muhtemel toksisitesini belirlemede önemli bir indikatör olarakkullanılabileceğini göstermiştir.
Abstract
The purpose of the present study was to investigate toxic effects of deltamethrin in Allium cepa L. For this purpose, we used parameters such as the root length, weight gain, germination percentage, frequency of micronucleus (MN) and chromosomal aberrations, anatomical changes with malondialdehyde (MDA) levels. Seeds were divided into 4 groups as 1 control and 3 application groups. The seeds of control group were treated with tap water only, and the seeds of application group were treated with 5, 10 ve 20 mg L -1 doses of deltamethrin for 72 h. The results showed that significantly reduced the, root length, weight gain and germination percentage, and the number of MN and chromosomal aberrations increased in all the application groups compared with results the control group. Chromosomal abnormalities were observed as sticky chromosome, bridge, fragment, c- mitosis and unequal distribution of chromatin. Also, results showed that deltamethrin at all doses caused anatomical damages such as flattened cell nuclei, necrosis, accumulation of some substances in the vascular tissue, thickening in the cortex cell walls, nonspecific vascular tissue and cell deformation. On the other hand, there was significant an alteration in the MDA levels of the root tips exposed to deltamethrin. Deltamethrin treatment significantly enhanced the lipid peroxidation and caused an increase in MDA levels in all treatment groups. The data obtained in this study showed that Allium cepa bioassays can be used as an important indicator to detect possible toxicity of deltamethrin.
