Dorystoechas hastata 'nın Kallus Kültürlerinin Oluşturulmasında Eksplant Tipi ve Bitki Büyüme Düzenleyicilerinin Etkisi

Özet
Bu çalışmanın amacı monotipik endemik Dorystoechas hastata bitkisinin kallus kültürlerinin oluşturulmasında eksplant tipi ve bitki büyüme düzenleyicilerinin etkisinin belirlemektir. Doğal ve in vitro elde edilmiş bitkilerin çeşitli kısımları tek başına BA veya 2,4-D ve kombinasyonlarını içeren ve hiçbir bitki büyume düzenleyici içermeyen 1/2 MS besi ortamlarına alınarak uygun eksplant tipi ve büyüme düzenleyici konsantrasyonu belirlenmiştir. Doğal ortamından toplanan yaprak, yaprak sapı ve in vitro fidelerden elde edilen yaprak sapı eksplantları kültüre cevap vermemiştir. In vitro fidelerden elde edilen yaprak eksplantları sadece 2 mgL-1 BA içeren 1/2 MS ortamında kallus oluşturmuşlardır. Kotiledon eksplantları ise kültüre farklı oranlarda kallus indüksiyon frekansı oluşturarak cevap vermişlerdir. En yüksek kallus indüksiyon frekansı 1 mgL-1 BA ve 2 mgL-1 2,4-D içeren 1/2 MS ortamında elde edilmiştir (% 55). Dorystoechas hastata bitkisinin kallus kültürlerinin oluşturulmasında kotiledonlar en uygun eksplant olarak belirlenmiştir
Abstract
This study was planned to determine the effect of explant type and plant growth regulators in the formation of callus cultures of the monotypic endemic Dorystoechas hastata plant. Various parts of the plants collected from their natural population and obtained in vitro were cultured on 1/2 MS medium containing BA or 2,4-D alone and combinations of these growth regulators. 1/2 MS medium without plant growth regulators was determined as control medium. The explants collected from wild and petiol explants obtained from in vitro seedlings did not respond to culture. Leaf explants obtained from in vitro seedlings formed calli only on 1/2 MS medium with 2 mgL-1 BA. Cotyledon explants obtained from in vitro seedlings were responded to the culture by creating callus induction frequency at different rates. The highest callus induction frequency was obtained on 1/2 MS medium with 1 mgL-1 BA and 2 mgL-1 2,4-D (55%). At the end of the experiments, cotyledons were determined as the most suitable explants in the formation of callus cultures of Dorystoechas hastata.
