Streptozotosin ile diabet oluşturulmuş sıçanlarda tiroid fonksiyonlarının incelenmesi

Özet
Bu çalışma streptozotosin (STZ) ile diabet oluşturulmuş sıçanlarda tiroid fonksiyonlarının incelenmesi amacıyla yapıldı. Bu amaçla Sprague-Dawley türü erkek sıçanlara 65 mg/kg olmak üzere tek doz STZ intraperitoneal olarak enjekte edildi. STZ enjeksiyonundan 48 saat sonra kan glukoz değerleri tayin edildi. Bu işlem 30 günlük periyot süresince haftada bir kez olmak üzere yapıldı. Bu süre içerisinde vücut ağırlıkları da takip edildi. 30 gün sonra diabetik sıçanların (n =20) ve kontrol grubunu oluşturan sıçanların (n =20) kalplerinden ponksiyonla kan alındı. Alman örneklerde serbest T3, total T s, serbest T4, total T4, ve insülin değerleri ölçüldü. Diabet oluşturulan grupta kontrol grubuna göre insülin düzeylerinde anlamlı azalma, kan glikoz düzeylerinde ise anlamlı bir artış saptandı. Yine diabetli grupta vücut ağırlıklarında anlamlı bir azalma görüldü. Abstract
Investigation of thyroid functions in streptozotocin-induced diabetic rats. In this study, the thyroid functions were investigated in streptozotocin-induced diabetic rats. Diabetes was induced by a single intraperitoneal injection of streptozotocin (65 mg/kg body weight) in male Sprague-Dawley rats. In order to decide whether diabetes had been developed or not, blood glucose values were determined 48 hours after the injection. The same procedure was applied once a week throught this 30 day period. 30 days after the injection, blood samples were drawn via the cardiac puncture from diabetic (n =20) and control rats (n =20), and serum concentrations of total T3, free T3, total 1, free T4, and insulin were measured. Serum insulin level and body weight were lower in the diabetic rats than in the control rats while serum glucose level was higher. Serum free T3, total T3, free T4 and total T4 levels were significantly reduced in the diabetic rats (Each of them p< 0.001).
