Özet

ÖZET. Karaciğerde Disse aralığında lokalize perisinüzoidal hücrelerin (Ito hücreleri, yağ-depolayan hücreler, stellat hücreler, liposit) ışık mikroskobik olarak gösterilmesinde kullanılan yöntemlerden birisi, rnodifıye altın impregnasyonudur. Ancak, yöntem 16 saat fiksasyondan sonra, frozen kesitlerin gold chloridde 16-24 saat impregnasyonu şeklindedir. Bu çalışmanın amacı; 16 saat ile sınırlı fıksasyon süresinin uzatılmasının, boyanma üzerine etkisini incelemektir. Çalışmada 16 saat, 24 saat, 3 gün, 7 gün, 15 gün ve 30 gün süre ile fiksatifte bırakılan erişkin Wistar albino, erkek sıçan karaciğerlerinden alınan frozen kesitlere altın impregnasyonu uygulandı. Elde edilen bulgulara göre; fıksasyon süresinin 3 güne kadar uzatılmasının boyanmayı olumsuz yönde etkilemediği sonucuna varıldı.

Abstract

SUMMARY. Modified gold impregnation is one of the methods that is used in light microscopical demonstration of perisinusoidal cells (Ito cells, fat-storing cells, stellate cells, lipocytes) that are localized in the space of Dis'se, in liver. In this method, frozen sections of tissues which are fixed for 16 hours previously, are impregnated in gold chloride solution for 16-24 hours. The aim of this study is to investigate the effect of longer fixation times on the staining results which is precisely determined as 16 hours. Liver specimens were taken from adult male Wistar albino rats and fixed for 16 hours, 24 hours, 3 days, 7 days, 15 days, and 30 days. After fixation, frozen sections of the specimens were cut and impregnated in gold chloride solution for 16-24 hours. According to the results, we concluded that prolonging the fixation time up to 3 days does not effect staining negatively.
