Özet

Lipopolisakkarit (LPS) tarafından oluşturulan endotokseminin patofizyolojik mekanizmalarının anlaşılmasın-da ve tedavisinde pek çok strateji geliştirilmesine rağmen, hala yoğun bakım ünitelerinin büyük bir problemi-dir. Bu çalışmada, endotoksik şok üzerine Apilarnilin koruyucu bir etkisi olup olmadığının, testis dokusundahistopatolojik değerlendirme ve DNA yapısındaki değişimler ortaya konularak belirlenmesi amaçlanmıştır.Çalışma için 64 adet Sprague-Dawley cinsi yetişkin erkek sıçan rastgele seçilerek 8 eşit gruba ayrılmıştır.Testis dokusunda rutin hematoksilen eozin boyaması ile histopatolojik değerlendirme yapılmış, komet testiile DNA yapısındaki değişiklikler, 6. saatin sonunda kontrol grubuyla karşılaştırılmıştır. Histolojik değerlendir-me sonucunda artan dozlarda apilarnil uygulanan gruplardan alınan testislere ait kesitlerde kontrol grubuile benzer şekilde histolojik yapı gösterdiği gözlenirken, LPS ve artan dozlarda apilarnil uygulanan gruplaraait testis dokusunda artan dozla birlikte lümendeki sperm miktarının arttığı görülmektedir. Buna ek olarak,LPS grubu ile karşılaştırıldığında bu gruplara ait seminifer tübüllerdeki hücreler arası bağlantı yerlerindedaha az ayrılmaların olduğu görülmektedir. DNA yapısındaki değişimler değerlendirildiğinde; LPS, uygulanangrupta kuyruk DNA yüzdesinde, kuyruk uzunluğunda ve kuyruk momentinde istatistiksel olarak artışa nedenolmuştur. LPS+Apilarnil'in verilmesi DNA hasarında azalmaya neden olmuştur. Apilarnil’in artan dozları DNAyüzdesi ve kuyruk uzunluğunda istatistiksel olarak azalmaya neden olmuştur. Sonuç olarak, apilarnil artandozlarda uygulanmasının, LPS'nin neden olduğu testiküler hasarı azaltığını ve bu etkinin de apilarnilin anti-oksidan kapasitesinden kaynaklandığını düşünmekteyiz.

Abstract

Although many strategies have been developed in the understanding and treatment of the pathophysiological mechanisms of endotexemia by lipopolysaccharide (LPS), it is still a major problem of intensive care units. In this study, we aimed to determine whether apilarnil has a protective effect on endotoxic shock by revealing histopathological evaluation and changes in DNA structure in testes tissue. For study, 64 Sprague-Dawley type adult male rats were randomly assigned to 8 equal groups. Histopathological evaluation was performed by routine hematoxylin eosin staining in testis tissue and changes in DNA structure were compared with the control group at the end of the 6th hour. As a result of histological evaluation, it was observed that sections of testis taken from increasing groups of apilarnil groups showed similar histological structure with the control group, and the amount of sperm in the lumen increased with increasing dose of testis tissue belonging to groups treated with apilarnil. In addition, there is less separation in the intercellular junction sites in the seminiferous tubules of these groups compared to the LPS group. When the changes in DNA structure were evaluated, LPS caused a statistically significant increase in tail DNA percentage, tail length and tail moment in the applied group. The administration of LPS+Apilarnil caused a decrease in DNA damage. Increased doses of apilarnil resulted a statistically significant reduction in DNA percentage and tail length. Increased doses of apilarnil resulted in a statistically significant reduction in DNA percentage and tail length. In conclusion, we think that apilarnil application of the increased doses decreases testicular damage caused by LPS and this effect is due to apilarnil antioxidant capacity.
