Deneysel serebral iskemi'de Lidokain'in etkileri

Özet
Bu çalışmada tek taraflı karotid arter ligasyonu ile oluşturulan deneysel hemisferal iskemi üzerine Lidokain'in etkisi araştırıldı, Halotan inhalasyonu ile anestetize edilen 20 farede sağ a. Carotis communis (ACCo) bağlandı. ACCo'nun ligasyonundan 30 dakika önce çalışma grubundaki 10 fareye 5 mg/kg'dan intravenöz olarak Lidokain, kontrol grubundaki 10 fareye aynı hacimde % 0,2 NaCI verildi. Ligasyondan 6 saat sonra her iki gruptaki denekler sakrifiye edilerek beyinleri çıkarıldı. Bu süre içerisinde deneklerdeki nörolojik değişiklikler araştırıldı, belirlenenler kaydedildi. Çıkarılan beyin örnekleri % 10'luk formaldehit içerisinde tesbit edildikten sonra, kesitler alınarak histolojik olarak değerlendirildi. Bu çalışmanın sonunda, istatistiksel olarak Lidokain ile tedavi edilen grupta meydana gelen iskemik nöronal değişikliklerin kontrol grubundan daha hafif olduğu görüldü. Buna dayanılarak Lidokain'in iskemide koruyucu etkisinin olduğu kanısına varıldı.
Abstract
This research presents the effect of Lidocaine after experimental hemispheral cerebral ischemia, induce by unilateral carotid artery ligation. Right common carotid artery ligation was applied to 20 mouses after being anesthetized by halothan inhalation. 10 mouses received lidocaine intravenously (5mg/kg), in the study group 30 minutes prior to carotid artery ligation, while the control group was receiving on equivalent volume of 5% dextrose, 0.2% saline. 20 mouses (10 lidocaine treated and 10 control) were sacrified after 6 hours of common carotid artery ligation and the brains were removed. Neurological changes were also observed within the duration of ischemia. After fixation, the brains were sectioned coronally into 3mm slices and the most extensive ischemic area for each subject was selected following histological assesment of the whole slices. After light microscopic evaluation of neural alternations in both lidocaine treated and control groups, we concluded that lidocaine has a significant protective effect in experimental ischemia.
