Özet

Amaç: Gıda kökenli yaşayabilen Enterococcus faecalishücreleri, tedavi esnasında yetersiz aseptik kontroldendolayı kök kanalını kontamine edebilir veya kanaltedavisinden sonra koronal sızıntı yoluyla kök kanaldolgusuna nüfuz edebilir. Enterokoklar yaygın olarakpeynir, et ürünleri, zeytin ve sebzeler gibi çiğ tüketilengıda türleri içinde veya üzerinde bulunur. Bu çalış-manın amacı bölge mizdeki 18 farklı peynir türündekiE.faecalis varlığını incelemektir.Gereç ve yöntem: Yerel süpermarketlerden teminedilen peynirlerden steril araçlar kullanılarak 1 gramlıkparçalar kesildi. Bu peynir parçaları 10 ml %2lik sod -yum sitrat buffer içerisinde bir dakika süre ile homo-jenize edildi. Daha sonra %2lik sodyum sitrat bufferile 10 kat seri dilüsyonları hazırlandı (10-1- 10-5). Her birdilüs - yondan 50 μL kanamisin eskülin azid agara(KAAA) ekim yapıldı ve 42oCde, %5 CO2 li ortamdainkübe edil di. Üç günlük inkübasyondan sonrakoloniler ince - lendi. İlk olarak eskülin hidrolizi sonucusiyah renkli koloni oluşturan bakteriler enterokokkolonileri olarak tanımlandı. Tipik enterokok kolonigörünümü olan ko- loniler KAAA kültür plaklarınasubkültür e edildi. Daha sonra Gram boyama ve katalaztesti ile incelendi ve Gram pozitif, katalaz negatifkoklar, üreticinin önerile - rine göre Vitek-2 otomatizesistemi ile tür düzeyinde tanımlandı.Bulgular: Bu peynir numunelerinin hiçbirinde yaşa -yabilen E.fae calis bulunmazken, 3 peynir numunesindeE.faecium , 2 peynir numunesinde E.durans, 1 peynirnumunesinde E.gallinarum,ve 2 peynir numunesindede Pediococcus spp barındırdığı gözlemlenmiştir.Sonuç: E.faecalis, özellikle Avrupadaki pek çok peynirtüründe g örülmektedir ve bu peynirlerdeki entero-koklar ile bunların oral kavitede bulunması arasındakiilişkiyi ortaya koyan çalışmalar yapılmıştır. Bu çalışma -da Türk peynirlerinde E.faecalise rastlanmamıştır, budurum kök kanallarında E.faecalis yüzdesini düş ürenbir neden olarak sunulabilir.

Abstract

Aim : Food -borne viable Enterococcus faecalis cellscontaminate the root canal during treatment due toinadequate aseptic control or pe netrate the root fillingvia coronal leakage after root canal treatment.Enterococci are commonly found in cheese, meatproducts, olives and vegetables. The aim of this studywas to investigate E.faecalis in 18 different cheese inCukurova region.Mat erials and method : Eighteen different types ofcheese were provided from local supermarkets andinvestigated for their enterococci content. 1 -g pieceswere cut out from the body of each cheese usingsterile instruments. The cheese pieces were homoge -nized for 1 min in 10 mL of a 2% sodium citrate bufferusing a tissue homogenizer. Subsequently, 10- foldserial dilutions were prepared in 2% sodium citratedown to 10-5. As much as 50 μL of each dilution wasplated on kanamycin aesculin azide agar (KAAA) usinga spiral diluter and incubated at 42 ºC in a 5% carbondioxide atmosphere. Colonies were counted after 3days of incubation. Enterococcus colonies werepresumptive ly identified by their blackening of theagar due to hydrolysis of esculin. Colonies that disp-layed the typical enterococcal growth and cell morp-hology were picked up and subcultured on KAAA pla -tes. Subsequently, a catalase test was performed andGra m -positive, catalase - negative cocci were identifiedto species level by Vitek- 2 automatized systemaccording to the instructions of the manufacturer.Result : None of the cheese samples contained viableE.faecalis, 3 cheese sample harbored E.faecium , 2che ese sample harbored E.durans , 1 cheese samplecontained E.gallinarum and 2 cheese sample harboredPediococcus spp.Conclusion: E.faecalis occur in a variety of cheeses,especially in Europe. And they find the relationenterococci in cheese and their occur ence in the oralcavity. We couldn t found E.faecalis in Turkish cheeseso it could be the reason of decreasing percentage ofE.faecalis in root canals.
