Özet

Bu araştırmada, ineklerde subklinik mastitislerin tanısında kulla¬nılan elektriksel iletkenlik (EC) yönteminin etkinliğinin, somatik hücre sayısı (SCC) ve California Mastitis Test (CMT) ile karşılaştırarak araştırılması a-maçlanmıştır. Çalışmada 3 farklı çiftlikte bulunan 49 ineğin toplam 183 meme lobundan alınan süt örnekleri materyal olarak kullanıldı. Süt örnekleri klinik : mastitis belirtisi göstermeyen ve laktasyonlarının ortalama 150. gününde bulu- nan ineklerden bir miktar süt sağıldıktan sonra alındı. Alınan süt örneklerine önce CMT'i uygulandı. Süt örneklerinin EC'leri Milk Checker cihazı (Eisai Co. Ltd.) kullanılarak saptandı ve somatik hücre sayıları kalitatif (nitel) sayım yön¬temi ile mikroskopik olarak yapıldı. Elde edilen sonuçlar variyans ve t-test yöntemine göre değerlendirildi. Toplam 183 süt örneğinin 50'si (%27.3) CMT (-), 40'ı (%21.8) CMT (+), 56'sı (%30.6) CMT (++) ve 37'si (%20.2) CMT (+++) olarak değerlendirildi. Ortalama elektriksel iletkenlik değerleri ise CMT (-) grubunda 5.41, CMT (+) grubunda 6.317, CMT (++) grubunda 6.867 ve CMT (+++) grubunda 7.85 mS/cm olarak belirlendi. California Mastitis Teşt sonuçlarına göre değerlendi¬rilen örneklerde SCC sayısı kalitatif olarak sırasıyla 100-200.000, 200-300.000, 300.000-1.000.000 ve 1.000.000'dan fazla olarak saptandı. Sonuç olarak subklinik mastitisli sütlerde elektriksel, iletkenliğin sap¬tanmasının CMT ve SCC ile benzerlik gösterdiği ve sahada kullanılabilir gü¬venli bir testolduğu ve subklinik mastitislerin teşhis yöntemi olarak kullanıla¬bileceği, diğer tanı yöntemleri ile kullanıldığında güvenilirliğinin daha da arta¬cağı kanısına varıldı.

Abstract

The aim of this study was to determine effectiveness of the method of electrical conductivity comparing somatic cell count and California mastitis test. In this study, the milk samples collected from 183 quarters of 49 cows in three different farms were used as the material. The samples were taken from the cows which not showing the signs of mastitis and at about 150th day of their lactation. The milk samples were taken during milking in the morning, after a few mililitres milk discarded. Firstly, CMT was applied, to the samples. The conductivities of the samples were determined by using Milk Checker. Somatic cell counts of the samples were carried out microscopically by using qualitative count method. All the results obtained have been evaluated according to variance and, t-test method. Of the 183 milk samples, 50 (27.3%) were CMT (-), 40 (21.8%) were CMT (+), 56 (30.6%) were CMT (++) and 37 (20.2%) were CMT (+ + +). The mean conductivity andSCC were 5.41, 6.317, 6.867, 7.85 mS/ml and 100-200.000, 200-300.000, 300.000-1.000.000 and above 1.000.000 cell/ml respectively in the groups of CMT (-), CMT (+), CMT (++) and CMT (+ ++). It has been reached the conclusion that the establishment of conductivity is safe and practical than CMT and SCC methods on the diagnosis of subclinical mastitis and it is also a cheap method, so as to use under farm conditions on the diagnosis of subclinical mastitis. Furthermore its safety would increase more when used together with the other methods.
