Bu çalışma, deneysel olarak yapılan pastırmaların mikroflorasında bulunan mikrokok ve stafilokokların klasik metot ve ticari API-ID 32 Staph sistemi ile karşılaştırmalı identifıkasyonu amacıyla yapıldı. Örneklerden izole edilen toplam 77 izolattan 75 'i (97.4) klasik yöntemle identifiye edilirken, bunlardan 55'i (% 73.3) stafilokok, 20'si (% 26.6) mikrokok olarak saptanmış, 2 izolat identifiye edilememiştir. API-ID 32 Staph sistemiyle ise, izolatların 53 'ü (% 70.6) stafilokok, 11'i (% 14.6) mikrokok olarak tanımlanırken, 11 izolat tanımlanamamıştır. Ayrıca S. saprophyticus, S. xylosus ve M. varians türlerinin her iki yöntemle de predominant olduğu, S. epidermidis, S. simulans, S. intermedius, S,: auricularis ile M. luteus, M. nishinomiyaensis, M. lylae ve M. roseus'un mikroflorada temsil edildiği saptanmıştır. Klasik yöntemle identifikasyonu yapılan mikrokok ve stafllokoklann, API-ID 32 Staph sistemi ile tiplendirilmesi sonucunda, 54 izblat (% 72) her iki yöntemle de benzer şekilde tanımlanırken, 21 izolatın (% 28) tanımlanmasında farklılık saptanmıştır. Farklılık gösteren izolatlardan l'i (% 1.3) soy. 9 'ıı (% 12) tür düzeyinde bulunurken, 11 'i (% 14.6) tanımlanamamıştır. Sonuç olarak, iki yöntem arasındaki farklılığın giderilebilmesi ve hızlı teknikle doğru tanımlama oranının arttırılabilmesi için, API-ID 32 Staph sistemine gıda hijyeni ve teknolojisi açısından önem taşıyan türlerin daha güvenli identifıkasyonunu sağlayacak bazı testlerin ilave edilmesi önerilir.

Abstract

The aim of this study is the comparative identification of micrococci and staphylococci present in the microflora of pastrami manufactured experimentally by using conventional method and the commercial system of API-ID 32 Staph. Seventyfive (97.4 %) of 77 isolates from pastrami samples were identified by conventional method, which 55 (73.3 %) of them were staphylococci and the rest 20 (26.6 %) were micrococci, but 2 isolates could not be identified. By using API-ID 32 Staph system, 53 isolates (70.6 %) were determined as staphylococci and 11 (14.6 %) were as micrococci, but 11 of isolates could not be identified. In addition to that, it was found that S.saprophyticus, S. xylosus and M. varians were revealed as predominant species by using both methods, and S. epidermidis, S. simulans, S. intermedius, M. luteus, M. nishinomiyaensis, M. lylae and M. roseus existed in microflora. At the result, the identification of 54 isolates (72 %) were similar while there is differents in determining 21 isolates (28 %) by both methods. One of different isolates (1.3 %) was found in genus level and 9 of them (12 %) were in species level, but 11 (14.6 %) could not be identified. As a consequence, in order that the difference between two methods could be recovered and the frequency for the precise identification could be increased, it might be recommended that some tests, which could provide more reliable identification of important species with respect to food hygine and technology be added to the system of API-ID 32 Staph.
