Özet

Bu çalışma, Türk salamura beyaz peynirinin yapımı ve olgunlaşması sırasında E. coli 0157 :H7'nin canlı kalma süresinin belirlenerek, potansiyel halk sağlığı riski oluşturup oluşturmadığını saptamak amacıyla yapıldı. Bu amaçla, pastörize süte (72°C, 2 dak) $10^2$ kob/ml, $10^4$ kob/ml ve $10^6$ kob/ml düzeylerinde E. coli O157:H7 inokule edildi. Starter kültür olarak, % 2 oranında Chr.Hansen R 707 peynir starter kültürü (Lactococcus lactis subsp. lactis , Lactococcus lactis subsp. cremoris) ile % 0.5 oranında Chr.Hansen YC 350 yoğurt starter kültürü (Streptococcus salivarius subsp. thermophilus, Lactobacillus delbrueckii subsp. bulgaricus) kullanıldı. Bütün peynir grupları 4°C'de, % 13 NaCl içeren salamurada, 120 gün muhafaza edildi. E. cali O157:H7'nin canlı kalma süresini belirlemek için sütten, telemeden ve olgunlaşmanın 1., 7., 15., 30., 60., 90. ve 120. günlerinde peynirlerden örnekler alındı. Tüm inokulasyon düzeylerinde teleme aşamasında ve 1. gün 1 log'lık bir artış saptandı. Olgunlaşmanın 90. günü I02 kob/ml inokulasyon düzeyinde, E. coli OI57:H7 sayısı 0.7 MPN/g'a, $10^4$ kob/ml düzeyinde, 4 MPN/g'a, $10^6$ kob/ml düzeyinde ise $5.3 x 10^1$ MPN/g'a düşmüştür. 120. Gün 10: kob/ml ve 104 kob/ml düzeylerinde inokulasyon yapılan peynir gruplarında E. coli 0157:117 tespit edilemezken, $10^6$ kob/ml düzeyinde inokulasyon yapılan peynir gruplarında, ortalama $1.5 x 10^1$ MPN/g E.coli 0157:H7 saptanmıştır. Sonuç olarak, E. coli O157:H7 ile kontamine olan Türk salamura beyaz, peynirinin, enfeksiyonun insanlara geçişinde önemli bir aracı olduğu ve potansiyel sağlık riski taşıdığı görüşüne varılmıştır.

Abstract

This study was conducted to determine the survival of E. coli 0157:117 during the cheese manufacturing and ripening periods, thus to indicate a potential public health hazard. For this purpose, E. coli 01577:H7 was inoculated at the level of $10^2, 10^4$ and $10^6$ cfu/ml to pasteurized milk (72°C for 2 min). Chr.Hansen R 707 cheese starter culture (Lactococcus lactis subsp. lactis , Lactococcus lactis subsp. cremoris) and Chr.Hansen YC 350 Yoghurt starter culture (Streptococcus salivarius subsp. thermophilus, Lactobacillus delbrueckii subsp. bulgaricus ) were added to the pasteurized milk at the rate of 2 % and 0.5 %, respectively. All cheese samples were ripened for 120 days in 13 % NaCl at 4°C. The samples were taken from milk, curd and the 1st, 7th, 15th, 30th, 60th, 90th and 120th days of ripening period of cheese and analysed for the numbers of E. coli 0157:H7. The population of E. coli 0157:H7 increased 1 log cfu/g in curd and first day of manufacturing with all of the inoculated levels. In the 90 dav of ripening period, E. coli 0157:H7 decreased from 102 cfu/ml to 0.7 MPN/g; from 104 cfu/ml to 4 MPN/g and from, 106 cfu/ml to 5.3x10' MPN/g in cheese samples. After 120 days, E. coli 0157:H7 was not detected in cheese samples, inoculated with the level $10^2$ cfu/ml and $10^4$ cfu/ml; on the other hand, it was detected at $1.5x10^1$ MPN/g in cheese samples inoculated with the level $10^6$ cfu/ml (pH 4.6, NaCl (m/m) : % 6.3±0.2). In conclusion, it was decided that, Turkish white brined cheese contaminated with E. coli 0157:117, could have a potential public health risk and serve as an important vehicle of transmission of infection to man.
