Özet

Bu çalışmada, makroskobik olarak kazeöz lenfadenitis tanısı konulan kavunlardan alman 41 adet lenf düğümünün mikrobiyolojik muayenesi amaçlandı. Alman materyaller % 7 koyun kanı içeren Brain Heart Infusion (BHI) agar ve % 7 defibrine at kanlı ağara iki seri olarak ekildi. Ekim yapılan petrilerin bir serisi mikroaerobik diğeri ise aerobik şartlarda olmak üzere 5 gün süre ile inkube edildi. Inkubasyona kaldırılan kültürler her gün kontrol edilerek, üreyen koloniler ınakroskopik ve mikroskopik açıdan incelenerek, biyokimyasal özelliklerine göre identifiye edildi. İncelenen toplam 41 örneğin 19 (% 46.3)'undan Corynebacterium pseudotıtberculosis ürerken, 8 (% 19.5) örnekte Micrococcus spp. üredi. Micrococcus spp. 5 örnekte saf kültür olarak, 3 örnekte ise C. pseudotuberculosis ile birlikte izole edildi. Ayrıca 3 (% 7.3) örnekten Staphylococcus aureus, 2 (% 4.S) örnekten Streptococcus epidermidis ve 3 ■(% 7.3) örnekten Pseudomanas aeruginosa izole edildi. S. aureus 2 (% 4.8) ve P. aeruginosa 1 (% 2.4) örnekte saf kültür olarak saptandı. izolasyon yapılan örneklerde C. pseudotuberculosis en yüksek oranda (% 46.3) bulunmuş, ayrıca örneklerden Micrococcus türlerinin de yüksek oranda (% 19.5) izole edilmesi, kazeöz lenfadenitis olgularında bu etkenlerin de önemini göstermektedir.

Abstract

Caseous lymphadenitis in sheep and goats is an infectious chronic disease caused by Corynebacterium pseudotuberculosis. The disease is characterized by encapsulated abscesses in the peripheral lymph nodes and sometimes lungs and other visceral organs. In this work a microbiological investigation was done on samples collected from sheep with clinical caseous lymphadenitis. In the study a total of 41 lymph nodes collected from 6 different regions were microbiologically examined. The samples, collected under sterile conditions were brought to the laboratory under the cold chain for examination. The contents of the lymph nodes were collected using a sterile syringe and the walls of the lymph were brushed with a sterile swab. The samples were then passed onto Brain Heart Infusion agar containing 7 % sheep blood. Two cultural conditions (aerobic and microaerobic) were used for each sample. The plates were incubated for 5 days and observed everyday for colony growth. The macroscopic and microscopic morphologies of the colonies were examined . The colonies were identified upon their biochemical characteristics. Of the 41 samples, 35 (85.4 %) were positive microbiologically. Corynebacterium pseudotuberculosis was isolated from 19 (46.3 %) of the 41 samples while Micrococcus spp. were grown in 8 (19.5 %) samples. Pure culture of Micrococus spp. were recovered from 5 samples whereas the other 3 samples had both Micrococcus spp. and C. pseudotuberculosis. In addition Staphylococcus aureus was isolated from 3 (7.3 %) samples, Streptococcus epidermidis from 2 (4.8 °/() and Pseudomanas aeruginosa from 3 (7.3 %) samples. Pure cultures of S. aureus were recovered from 2 (4.8 %) samples and a pure culture of P. aeruginosa form 1 (2.4 %) sample. The most commonly isolated organisms from the samples examined were C.pseudotuberculosis (46.3 %) and Micrococcus spp. (19.5 %). These results indicated the significances of these two microorganisms in the etiology of caseous lymphadenitis.
