Bromokriptin ve GnRH'nın laktasyondaki ratlarda serum prolaktin ve LH konsantrasyonları üzerine etkisi

Özet
Sunulan çalışmanın amacı, laktasyon sürecindeki ratlarda GnRH ve bromokriptin uygulamalarının serum LH ve PRL düzeylerine etkisini araştırmaktır. Çalışmada materyal olarak postpartum 6-11. günler arasında bulunan 28 Sprague-Dawley ırkı rat (n=28) kullanıldı. Ratlar her grupta 7 anne bulunan (n=7) 4 gruba ayrıldı. Grup I (n=7) kontrol grubu olarak ayrıldı ve intravenöz yolla serum fizyolojik uygulandı. Grup H'de bulunan ratlara (n=7), 25 ng GnRH, Grup IH'e (n=7) 500 Hg bromokriptin ve Grup IV'e (n=7) aynı dozlarda GnRH + bromokriptin intravenöz yolla uygulandı. Yavrularından 6 saat süre ile ayrı tutulan annelerin A. carotis communis'lerine kanül yerleştirildi ve bu yolla emzirme öncesi 10. dakikada ve hemen sonrasında kan alındı, ilaç uygulamalarını izleyen 15, 30, 45 ve 60. dakikalarda kan alımına devam edildi. Serum PRL ve LH ölçümleri EIA yöntemine göre yapıldı. Kontrol grubunda emzirme esnasında serum PRL konsantrasyonu 58.7± 9.7 $mu$ g/l ile 63.7±13.2 $mu$ g/1 arasında değişkenlik gösterdi. On beşinci dakikada PRL'nin 692.3±67.1 $mu$ g/l seviyesine çıktığı gözlendi. Serum LH konsantrasyonu ise emzirme süresince 18.4±3.1 $mu$ g/l ile 23.6±4.2 $mu$ g/l seviyeleri arasında seyretti ve emzirmeyi izleyen dönemde (15. dakika) 6.1±0.9 $mu$ g/l'den 60. dakikada 4.8±0.6 $mu$ g/l seviyesine indi. GnRH uygulamasının (Grup II) PRL konsantrasyonu üzerindeki etkisi oldukça belirgin ve önemli oranda bulundu. Serum LH konsantrasyonunun uygulama öncesi 22.5±3.9 $mu$ g/l olan seviyesi 15., 30. ve 45. dakikalarda artarak sırasıyla 33.7±2.5 $mu$ g/l, 40.1±4.3 $mu$ g/l, 51.4±4.9 $mu$ g/l'ye çıktı ve 60. dakikada 38.9±7.2 $mu$ g/l'ye düşüş gösterdi. Bromokriptin uygulaması (Grup III) prolaktin salınımını uygulama anında 48.5±3.6 $mu$ g/l'den 60. dakikada 13.4±4.3 $mu$ g/l'ye düşürdü. Aynı şekilde, LH salınımında da düşüş saptandı. GnRH+bromokriptin (Grup IV) PRL konsantrasyonunu 0. dakikada 52.9±7.1 $mu$ g/l'den 15. dakikada 23.1±1.6 $mu$ g/l'ye 45. dakikada 13.5±5.2 $mu$ g/l'ye düşürürken, serum LH konsantrasyonunda da belirgin bir artışa neden oldu. Sonuç olarak, emme stimulusu ve PRL'nin laktasyon sürecinde gonadotropin şahmınım baskıladığı, bromokriptin'in tek başına veya GnRH ile birlikte kombine uygulamaları sonucunda serum PRL konsantrasyonunun azaldığı, dolayısı ile bu etkinin ratlarda galaktopoezisin sonlandırılmasında kullanılabileceği kanısına varıldı.
Abstract
The aim of the study to detect the effectiveness of exogenous GnRH and bromocryptine administration during the nursing period on serum prolactin and luteinizing hormone concentrations. Twenty-eight Sprague-Dawley rats being in postpartal 6-11 days were used as materials. Rats were divided into 4.groups each contained 7 mothers. Seven rats which were in control group (Group I) only have received saline solution via intravenously. 25 ng gonadorelin (Ovarelin®, DlF) to Group II (n=7), 500 (jg bromocryptine (Parlodel®, Sandoz) to Group III (n=7) and GnRH+bromocryptine to rats in Group IV (n=7) were applied via intravenously. Mothers separated from their pups for 6 hours and at the end of this period, the pups have been come together with their mothers for them to suck for 10 minutes and separated again. Blood samples were collected at the beginning of nursing (-K)-min) and 10 minutes after (0-min). Immediately the second sample were collected drugs were injected. Blood was withdrawn through the cannula at 15, 30, 45 and 60 min after all the pups had separated. PRL and LH concentrations were measured by EIA method. In control group (Group I), PRL concentration was fluctuating between 58.7± 9.7 $mu$ g/l - 63.7±13.2 $mu$ g/l at -10-min and 0-min, respectively. It was observed that PRL rose sharply to 692.3±67.1 $mu$g/l at 15 min. During the nursing period LH concentration was between 18.4±3.1 $mu$ g/l - 23.6±4.2 $mu$ g/l. LH levels were 6.1±0.9 $mu$ g/l at 15-min and remained suppression throughout the 60-min (4.8±0.6 $mu$ g/l). In Group II, GnRH has clearly stimulated the increase of PRL release. LH concentration was 22.5±3.9 $mu$ g/l during the nursing period. After the injection of GnRH, LH levels was elevated at 15, 30 and 45 minutes to 33.7±2.5mg/l, 40. l±4.3 $mu$ g/ and 51.4±4.9 $mu$ g/l, respectively and1 slight decrease shown at 60-min to 38.9±7.2 $mu$ g/l. Bromocryptine (Group III) induced a prompt suppression of PRL from 48.5±3.6 $mu$ g/l at the injection time to 13.4 ±4.3 $mu$ g/l at 60-min. LH release was similarly decrease in response to bromocryptine. Combined bromocryptine and GnRH treatment (Group IV) suppressed the PRL rise from 52.9±7.1 $mu$ g/l at 0-min to 23.1±1.6 $mu$ g/l at 15-min and 13.5±5.2 $mu$ g/l at 45-min, respectively. However, LH concentration was clearly elevated in this group. As a consequent, it,has been established that prolactin and suckling stimulation inhibit gonadotrophin secretion during lactation period and administration of the bromocryptine alone or combined with GnRH may terminate the galactopoiesis in the rats, as a result of decreased prolactin levels.
