Türkiye'de maedi-visna enfeksiyonunun seroepidemiyolojisi ve virus izolasyonu

Özet
Maedi-visna, klinik ve patolojik olarak iki ayrı formda gözlenen ve inktıbasyon süresinin uzun olması nedeniyle yavaş seyirli virüs enfeksiyonları içerisinde yer alan, koyunların viral bir enfeksiyonudur. Enfeksiyonun Türkiye'de varlığı yapılan çalışmalar ile daha önce ortaya konulmuştur. Bu araştırmada ise, maedi-visna enfeksiyonunun varlığı bilinen işletmelerde bulunan koyunlar örneklenerek, enfeksiyonun takibinin yapılması ve maedi-visna virüsünün humoral ve hücresel immun yanıtın varlığına rağmen hayvanlarda persiste kalması nedeniyle, virusa spesifik antikor saptanan hayvanlardan virus izolasyonu amaçlanmıştır. Bu amaçla, kamuya ait 10 koyun yetiştiriciliği işletmesinde bulunan 628 koyundan kan örneği sağlanmıştır. Maedi-visna antikoru yönünden AGID testi ile kontrol edilen 628 koyundan 168'inde (%26.7), örneklenen işletmelerden 9'ıında (%90) enfeksiyonun varlığı saptanmıştır. Maedi-visna antikoru saptanan koyunlara ait löykosit örneklerinden virüs izolasyonu amacıyla koyun choroid plexus (SCP) hücre kültürü kullanılmış vp 2 koyuna ait löykosit örneğinde, maedi-visna virüsü için spesifik olan çok çekirdekli dev hücresi oluşumuyla karakterize sitopatik etki (CPE) oluşumu tespit edilmiş olup, izolatların indirekt immunfloresan testi ile identifikasyonu yapılmıştır.
Abstract
Maedi-visna (MV) is an important virus infection of sheep having prolonged incubation period (slow disease) and reflecting two distinct forms clinically and pathologically. Presence of the infection in Turkey have been shown by previous researches. In this study, sheep flocks which were proven to be infected with MV virus previously were sampled in order to screen the present status of infection and to isolate the causative agent from seropositive individuals, since the virus might be persistent in such animals instead of affective humoral and cellular immunity. For this purpose, the blood samples were taken from 628 sheep in 10 flocks. In 168 (26.7%) of the serum samples, MV-virus specific antibodies were detected by agar gel immunodiffusion (AGID) assay and presence of the infection were verified in 9 of the flocks sampled. For virus isolation attempts, leukocyte samples obtained from seropositive sheep were inoculated into SCP cell line. In two SCP cultures, lentivirus specific cytopathic changes characterised by formation of multinuclear giant cells were observed. Identification of these two isolates was confirmed by indirect im-munofluorescence test.
