Quantitative comparison of various methods and solutions for detecting eggs of anoplocephala perfoliata in horse faeces

Özet

At dışkısında Anoplocephala perfoliata yumurtalarını saptamak amacıyla sedimentasyon ile sedimentasyon+santrifüj-flotasyon yöntemlerinin karşılaştırılması yapılmıştır. Nekropside A.perfoliata taşımadığı belirlenen atlara ait dışkının 10'ar gramına 5, 10, 25, 50, 100 ve 200 yumurta eklenmiştir. Dışkı örnekleri sedimentasyon ve üç farklı solüsyon (NaCl+$ZnCl_2$ d. 1 .3, $ZnSO_4$ d: 1.3 ve doymuş şekerli su d: 1.3) kullanılarak sedimentasyon+ santrifüj-flotasyon yöntemleriyle veya yöntem tekrarlanmaları ile incelenmiştir. En fazla yumurta, iki defa tekrarlanan sedimentasyon ile doymuş şekerli su solusyonuyla yapılan sedimentasyon+santrifüj-flotasyon yöntemlerinde bulunmuştur.

Abstract

Sedimentation and sedimentation+centrifugation-flotation methods for recovery eggs of A.perfoliata from horse faeces were tested for efficiency and suitability for routine use. Faecal samples were collected from necropsied horses without A. per-foliata infected. The tests were done under standardised conditions on 10 g of horse faeces contaminated experimentally with 5, 10, 25, 5Q, 100 and 200 eggs of A. perfoliata. The samples were examined using sedimentation and sedimentation+centrifugation-flotation methods to use three different flotation solutions (NaCl+$ZnCl_2$ d:1.3, $ZnSO_4$ d:1.3 and saturated sucrose solution d:1.3). Significant best rate of recovery was achieved using sucrose solution for sedimentation+centrifugation-flotation method and double sedimentation.
