Gebe ineklerde uygulanan aşıların kolostrum ve buzağıda IgG konsantrasyonu üzerine etkileri

Özet
Bu çalışmada, ineklerde gebeliğin ileri dönemlerinde uygulanan aşıların kolostrum ve buzağı kan serumundaki IgG seviyesi üzerine etkisi araştırıldı. Araştırmada hayvan materyalini 40 baş Holstein ırkı inek ve bunlara ait 40 baş buzağı oluşturdu. Seçilen hayvanlar 20 baş uygulama ve 20 baş kontrol olmak üzere 2 gruba ayrıldı. Uygulama grubundaki inekler, inaktif Rota ve Coronavirus antijenleri ile E. coli bakteri toksoidini içeren aşı (ScourGuard 3(K)) ile, gebeliğin son 1,5 ayında, 2 hafta arayla 2 kez kas içi yolla aşılandı. Doğum yapan ineklerden, doğumdan hemen sonra 5 mi olmak üzere kolostrum alındı. Yeni doğan buzağıların ise yeterli kolostrum alması sağlandı ve doğumdan 24 saat sonra vena jugularisten 10 mi kan toplandı. Kan örnekleri 3000 devir/dakika hızında 10 dakika süresince santrifüj edildi ve serumları çıkarıldı. Kolostrum ve serum örneklerinin IgG miktarı Single Radial Immunodiffusion (SRID) yöntemi ile belirlendi. Buzağıların immun yapısının ortaya konulması amacıyla, serum IgG seviyesine göre; IgG seviyesi 800 mg/dl veya daha az olanlar, 800 mg/dl ile 1600 mg/dl arasında bulunanlar ve 1600 mg/dl'den yüksek olanlar şeklinde 3 gruba ayrıldı, ineklerin kolostrumlan ise içerdikleri IgG konsantrasyonuna göre 3200 mg/dl'den altında, 3200-4800 mg/dl arasında ve 4800 mg/dl'den fazla olmak üzere yine 3 grupta değerlendirildi. Araştırma sonucunda, kontrol ve uygulama grubundaki ineklerin ortalama kolostrum IgG düzeyleri sırasıyla 3542±1368 mg/dl ve 5500±1204,3 mg/dl idi (P
Abstract
In this study, the effects of vaccination on the level of IgG in colostrum and calf blood was investigated when administered late periods of pregnancy in cows. The study materials consisted of 40 cows and 40 calves belonging to their. The cows were selected among clinically healty Holstein cows. Selected animals divided in to 2 groups; 20 cows and their calves control group and 20 cows and their calves study group. Cows were intramuscularly vaccinated with vaccines (ScourGuard 3(K)) containing inactive Rota and Coronavirus antigens and E. coli bacterin toxoid twice in 2 weeks on the last 1,5 month of pregnancy. Five ml of colostrum was collected, immediately after the cows gave birth. The calves were made sure they had enough colostrum and 10 ml of blood was collected from jugular vein after 24 hours from birth. Blood samples were santrifuged for 10 minutes with a speed of 3000 rpm and the serumen was separated. Later on, IgG level in serum and colostrum was determined with Single Radial Immunodiffusion (SRID) method. Calves were divided into 3 groups according to the serum IgG levels as follows; The IgG level less then 800 mg/dl, between 800-1600 mg/dl and more than 1600 mg/dl. Cows were divided into 3 groups according to the colostrum IgG levels as follows; Less than 3200 mg/dl, between 3200-4800 mg/dl and more than 4800 mg/dl.
