Modifiye akrilik ve yüzeylere candida albicans tutunmasının in vitro olarak araştırılması

Özet
Protez stomatitlerinin patogenezinde, kaidenin doku yüzeyinde çok sayıda kandidanın, özellikle de C. albicansların tutunması önemli bir etiyolojik faktör oluşturur. Bu çalışmada, akrilik rezinlerin yüzey ıslanabilirliğinin, C. albicans tutunması üzerine etkisi değerlendirilmiştir. Kontrol ve deney gruplarını oluşturan Poli(metilmetakrilat) örnekler, ısı ile polimerizasyon yöntemiyle hazırlandı. Yüzey ıslanabilirliğini değiştirmek amacıyla deney grubu örnekleri, oksijen gazı ile değişik boşalım güçleri altında glow discharge reaktöründe radyofrekans plazması ile modifiye edildi. Islanabilirlik özelliği, sesil damlacık yöntemi kullanılarak yapılan temas açısı ölçümleri ile belirlendi. Tutunma deneylerinde kullanılmak üzere C. albicans suşu seçildi ve kültür ortamında çoğaltıldı. Uyarılmamış insan tükürüğü ile muamele edilen akrilik örnekler, 37 $circ$ C de 2 saat boyunca kandida solüsyonunda inkübe edilerek alkol ile fikse edildi ve gram boyamaları yapıldı. Abstract
The important aethiologic factor in the pathogenesis of denture-induced stomatitis is the presence of numerous yeasts, usually C. albicans, on the fitting surface of the dentures. In the present study, the effect of surface wettability of acrylic resins on the adherence of C. albicans was evaluated. Poly(methylmethacrylate) samples including control and experimental groups were prepared by using heat compression mold technique. To create a hidrophobicity gradient, experimental groups were exposed to a radio frequency glow discharge in an O2 atmosphere under different discharge powers. To characterize the wetting properties, an expression of surface hidrophobicity, contact angle measurements were performed by the sessile drop method. The organism used was C. albicans (ID:32). Acrylic samples were coated with unstimulated whole saliva collected from a healthy man. The fungal suspension was poured onto saliva-inoculated samples and incubated at 37 $circ$C for 2 hours. The samples were then fixed with methanol and Gram stained. Adhered candidial cells were examined by light and scanning electron microscope.
