Ordu ilinde bulunan bazı Rubus (Rosaceae) türlerinde trnL-F gen bölgeleri ile filogenetik ilişkilerin belirlenmesi

Özet
Bu çalışmada; Ordu ilinde yaygın bulunan yabaniRubus türlerinin filogenetik farklılıkları trnL-F genbölgeleri kullanılarak belirlenmeye çalışılmıştır.Ordu’nun farklı noktalarından 24 adet Rubus spp.örneği toplanmıştır. Örneklerden DNA izolasyonuHaymes’in (1996) geliştirdiği izolasyon protokolüuygulanarak yapılmıştır. Örneklerin trnL-F genbölgesini belirlemek için, GenBank’tan alınanreferans diziler ile çalışmanın sekans dizilerikarşılaştırılmıştır. MEGA6 paket programıyardımıyla genetik uzaklıklar hesaplanmış vefilogeni ağacı oluşturulmuştur. Örneklerden 6haplotip sonucu çıkmıştır. Bunlar; R. sanctus(EF055343), R. silvaticus (KM036881), R. divaricatus(KM036924), R. conothyrsoides (KM036856), R.capricollensis (KM036923) ve Rubus sp. (KT581217)ile %100 nükleotid dizisi benzerliğiyle akraba olarakdeğerlendirilmiştir.
Abstract
In this study; the phylogenetic differences of some Rubus genotypes naturally grown in Ordu province in Black Sea Region were tried to be determined by using trnL-F gene regions. 24 Rubus spp. were collected with a survey. Haymes (1996) DNA isolation protocol was used with little modification to isolate the DNAs from the samples. In order to determine the trnL-F gene region of our samples, the reference sequences from GenBank were compared with the sequences obtained in our study. Genetic distances were calculated with the help of MEGA6 package program and phylogeny trees were drawn. The samples were categorized in 6 Haplotype. These; Haplotype 1 (F3, P1, P3, P4, U1, U2, O2, O3, O4, CM1), Haplotype 2 (F5), Haplotype 3 (P2), Haplotype 4 (P5), Haplotype 5 (S1, CM2, CM3), Haplotype 6 (O1, CT1, CT2, CT3), respectively, R. sanctus (EF055343), R. silvaticus (KM036881), R. divaricatus (KM036924), R. conothyrsoides (KM036856), R. capricollensis (KM036923) and Rubus sp. (KT581217).
