CD53 monoclonal antibody induces expression of CD35 and HLA-DR molecules on the U937 cell line

CD53 is a pan-leucocyte antigen which spans the plasma membrane four times and is a member of transmembrane 4 super family (TM4SF). The precise function of TM4SF is not known but data suggest a role for this family in the regulation of cell development, proliferation and activation. It has been shown that CD53 is involved in signal transduction and activa¬tion of monocytes and T cells. In this study modulation of expression of CD35 molecule, a transmembrane signal trans¬ducer, and HLA-DR on U937 cell line was analyzed after addition of a specific monoclonal antibody (mAb) raised against CD53 molecule into the culture. Addition of this mAb (designated as SU14) into the culture of U937 cell line results in an increase in the expression of CD35 and HLA-DR molecules. In a similar experiment of Jurkat cell line, a significant increase in the expression of HLA-DR and IL-2R was observed. These data indicate that SU14 mAb binding induces expression of activation markers on the surface of Jurkat cells. Binding of this mAb of Jurkat cells was also followed by an increase in the expression CD44 molecule. Turk J Haematol 1997; 14:93-96.
