Octreotide' nin stres uygulanan sıçanlarda mide lezyonları ve mide mukozal bariyeri üzerindeki etkileri

Özet

Soğuk + immobilizasyon stresi uygulanan sıçanlarda somatostatin analoğu sandostatinin mide lezyonları ve mide mukozal bariyeri üzerine etkilerini araştırmak amacı ile yapılan bu çalışmada 21 adet 150-200 gr ağırlığında Swiss Albino sıçan kullanıldı. Hayvanlara 24 saat açlıktan sonra 1.grub (n=7) stres uygulanmayan kontrol grubu olarak alındı. 2. gruba (n=7) soğuk + hareketsizlik stresi uygulandı. 3. gruba (n=7) stresten bir saat önce 100 µg/kg sandostatin (sandoz) subkutan olarak uygulandı. Daha sonra mideleri ülseratif lezyon yönünden incelenerek ülser indexi saptandı. Mide mukozal bariyeri komponentlerinden olan mukus ve fosfolipid miktarları Corne ve Baur metodu ile tespit edildi. Stres uygulanan ratlarda mide mukozal bariyerinin önemli komponenentlerinden olan mukus ve fosfolipid düzeyleri önemli bir şekilde azaldı (p<0.05, p<0.05). Sandostatinin mide lezyonlarını önlediği görüldü (p<0.01). Ayrıca sandostatin uygulanan sıçanlarda mukus ve fosfolipid miktarlarının stres uygulanan kontrol grubuna göre artmış olduğu saptandı (sırasıyla, p<0.05, p<0.005). Bulgularımıza göre stresin neden olduğu akut hemorajik mide lezyonlarının önlenmesinde ve mide mukozal bariyer parametleri üzerinde sandostatinin etkin olduğunu göstermektedir.

Abstract

The aim of this research was to investigate the effects of octreatide on gastric lesions and mucosal barrier in rats exposed cold +immobilisation stress. Twenty four Swiss Albino rats (approx 150-200 g) were used for this study. Feeds were witheld from to rats for 24 hours, after which they were divided in to three groups. Each group consisted of seven rats; 1 (n=7) non/stress-induced control group, 2 (n=7): cold and immobilisation stress was induced, 3 (n=7): 100 µg/kg octreatide was administered subcutaneously one hour prior to stress induction. The rats were then killed and their stomachs examined for ulcerative lesions and ulcer indexes considered. The amounts of mucus and phospholipid, important components of the gastric mucosal barrier, were then rated using Corn and Baur methods and were found to be significantly decreased (p<0.05, p>0.05) in rats exposed to stress. In the third group given sandostatin, it was found that gastric lesions had been prevented. Moreover, mucus and phospholipid levels were hig-her in this group than the stress exposed control group. (p<0.05, p<0.05 respectively). According to our findins, octreatide is effective in preventing acute hemorrhagic gastric lesions caused by stress and in maintaining gastric mucosal barrier paramters.
