Epidermal büyüme faktörünün mide mukozası lipid peroksidasyonu üzerindeki etkisi

Özet
Epidermal büyüme faktörünün (Epidermal Growth Factor, EGF) çeşitli ülserojenlere karşı mide mukozasını koruduğu bilinmektedir. Mide mukoza hasarında serbest oksijen radikalleri mediator olarak rol oynamaktadır. Bu çalışmada EGF'nin mide mukozasında malondialdehid (MDA) ve glutatyon (GSH) düzeyleri üzerindeki etkisi araştırıldı. Çalışmada, soğuk-immobilizasyon ile stres ülseri oluşturulan 50 ve sağlam 10 erişkin rat kullanıldı. Sağlam gruba stres uygulanmazken; stres uygulanan ratların birinci grubuna intragastrik EGF (30 $mu$g/kg, 7 gün süreyle), ikinci gruba intragastrik Serum Fizyolojik (SF), üçüncü gruba intraperitoneal EGF (30 $mu$g /kg, 7 gün süreyle), dördüncü gruba intraperitoneal SF verildi ve beşinci gruba ise tedavi verilmedi. EGF gerek hem intraperitoneal (i.p) hem de intragastrik (i.g.) yolla verildiğinde, SF ve kontrol grupları ile karşılaştırıldığında ülser alanlarını anlamlı olarak azaltmıştır. Stres uygulanan raflarda mide MDA düzeyleri sağlam gruptakine göre anlamlı olarak daha yüksek bulunurken, mide GSH düzeyleri açısından bir farklılık bulunamadı. Parenteral EGF uygulanan ratlarda mide MDA düzeyleri hem SF uygulanan ve hem de intragastrik EGF/SF uygulanan gruplardakine göre anlamlı olarak daha düşük bulundu (p<0.05). İntragastrik EGF verilen ratların mide mukozası MDA düzeyleri SF grubundakine göre daha düşük olmakla birlikte istatistiksel olarak anlamlı bulunmamıştır (p= 0.152). Mide mukozası GSH düzeyleri açısından tedavi grupları arasında anlamlı farklılık saptanmadı. Sonuç olarak EGF'nin, özellikle parenteral yolla verildiğinde, mide mukozasında serbest radikal düzeylerini azaltmada daha etkili olduğu stres ülserlerini önlemede de yararlı olabileceği kanaatine varılmıştır.
Abstract
Epidermal growth factor (EGF) is known to be a protective factor for gastric mucosa against various ulcerogenic agents. Lipid peroxidation products may have a role in the pathogenesis of mucosal damage. In this study, we investigated the effects of EGF on ulcer healing and on malondialdehyde (MDA), a major lipid peroxidation product, and glutathione (GSH) levels of gastric mucosa in a cold immobilization stress model. Fifty female adult Wistar rats, exposed to cold and immobilization stress, and 10 non stress exposed rats were included in the study. The rats exposed to stress were divided into five groups: intragastric EGF (30 $mu$g/kg/d, i.g., 7 days), intragastric normal saline (SF), intraperitoneal EGF (30 $mu$g/kg/d, i.f., 7 days), intraperitoneal SF and no treatment. Both the i.p.EGF and i.g. EGF groups significantly decreased the ulcer area of gastric mucosa when compared to the control and SF groups. Gastric MDA levels were significantly higher in stress groups compared to the control group. However, gastric GSH levels were similar in all groups. Gastric MDA levels were found to be significantly lower in the i.p EGF group compared to the i.p. SF, intragastric EGF and SF groups (p<0.05). The gastric MDA levels of the i.g. EGF group were found to be slightly lower than that of the i.g. SF, but this difference was not statistically significant (p=0.152). There was no significant difference in the gastric GSH levels of treatment groups exposed to stress. It is concluded that EGF, especially intraperitoneal, may prevent stress ulcers by means of decreasing the levels of lipid peroxidation products of gastric mucosa.
