Effect of Zn, Fe and Cu Content on Phytochemical Investigations and Antimicrobial Potential of Alternanthera brasiliana (L.) O. Kuntze Leaf Extracts Procured From Two Different States of India

Hindistan'ın iki farklı bölgesinden temin edilen Alternanthera brasiliana (AB) bitkisinin üç farklı yaprak ekstrelerinin çinko (Zn), demir (Fe) ve bakır (Cu) içeriğinin, bitkinin fitokimyasal bileşimi ve patojenik bakterilere karşı potansiyel antibakteriyel aktivitesi üzerindeki etkisi araştırılmıştır. Gölgede kurutulmuş yaprak örneklerindeki Zn, Fe ve Cu miktarları atomik absorbsiyon spektrofotometresi (AAS) kullanılarak belirlenmiştir. Su, metanol ve su:metanol (1:1) karışımı ile hazırlanan ekstrelerin mikrobiyosit potansiyel agar difüzyon tekniği kullanılarak değerlendirilmiştir. Ampisilin (30µg/mL) standart olarak kullanılmıştır. Ekstrelerin Minimum İnhibisyon Konsantrasyonu (MIC) seri dilüsyon metoduyla, mikrobiyal aktivitenin inhibisyonu agar dilüsyon yöntemiyle değerlendirilmiştir. Batı Bengal örneklerindeki Zn, Fe ve Cu miktarlarının daha yüksek olduğu belirlenmiştir. Bununla beraber, doz bağımlı (**P<0.05) antimikrobiyal etki tespit edilmiştir. 200 mg/mL dozda Batı Bengal'den elde edilen AB'nin sulu ve metanollü ekstresi (sırasıyla ABAE ve ABME), gram pozitif bakterilere karşı anlamlı derecede etkili bulunmuş, maksimum inhibisyon zonu Straphyloccus aureus (13.0 ± 0.011**)'a karşı kaydedilmiştir. Bu varyasyonun bitkinin coğrafik lokasyonu, toprağın pH'sı, elementlerin miktarı ve çözücü seçiminden kaynaklı olabileceği düşünülmüştür
Abstract
The effect of zinc (Zn), iron (Fe) and copper (Cu) on phytoconstituent content and the potential antibacterial activity of three different leaf extracts of Alternanthera brasiliana (AB) against pathogenic bacteria, procured from two different states of India, was evaluated. Shade dried leaf samples were separately determined for Zn, Fe and Cu content through atomic absorption spectrophotometer (AAS). Further extraction was carried out using aqueous, methanol and mixture of aqueous and methanol (1:1) and microbioside potential were evaluated by agar well diffusion technique. Ampicillin (30µg/mL) was used as standard. The Minimum Inhibition Concentration (MIC) of all the extracts was carried out by serial dilution method and thereafter inhibitory microbial activity was carried out by agar plate method. The amount of Zn, Fe and Cu were higher in West Bengal sample. Furthermore, the concentration dependent (**P<0.05) potential antimicrobial activity was observed. At the dose of 200 mg/mL, combined aqueous and methanol extract of AB (ABAE and ABME respectively) from West Bengal, gave significant results against gram positive bacteria where maximum zone of inhibition was recorded against Straphyloccus aureus (13.0 ± 0.011**). Such variation may be due to the geographical location of the plant, soil pH, content of elements and on the effects of choice of solvent
