Özet
Bu çal›flmada Veteriner Hekimlikte ve tar›msal alanlarda kullan›lan baz› kimyasallar›n (Azametifos, Diklorvos, Metil parathion) mutajenik etkileri Somatik Mutasyon ve Rekombinasyon Testi (SMART) ile araflt›r›ld›. SMART yöntemi iflaret genleri tafl›yan heterezigot sinek kanatlar›n›n hücre fenotipi üzerine kimyasallar›n etkisini gözlemek ilkesine dayan›yordu. Kullan›lan kimyasallar›n letal dozlar› belirlendi. Toplam mutasyon ve mutasyon gözlenen kanat say›s› aras›nda pozitif bir korelasyon gözlendi. Ayr›ca gözlenen mutasyonlar, kanat bafl›na mutasyona u¤ram›fl hücrelerin oluflturdu¤u kümelerin say›s›na ve mutasyon tiplerine göre s›n›fland›r›ld› ve istatistiksel analizler yap›ld›. Kullan›lan kimyasallar›n genotoksik etkilerine göre Diklorvos, Metil parathion ve Azametifos fleklinde s›raland›¤› belirlendi. Bu çal›flma, pestisidlerin insan ve çevre sa¤l›¤› için oluflturaca¤› zararl› etkilerden korunmak için, pestisid kullan›m›n›n elverdi¤ince aza indirgenmesinin gerekli¤ini ve e¤er kullan›m zorunlu ise pestisidlerin do¤ru kullan›m›n›n çok önemli oldu¤unu göstermektedir.
Abstract
In this study mutagenic effects of some chemicals used in Veterinary Medicine and agricultural fields was investigated by Somatic Mutation and Recombination Test (SMART). SMART test was to observe the chemical effects on wing phenotype of the trans- heterozygote flies carrying marker gene. Lethal doses of the chemicals used were determined. A positive correlation was observed between total mutation and the number of wings having mutations. In addition, the observed mutations were classified according to the size and the type of the mutations per wing. Chemicals used were ranked as Dichlorvos, Methyl parathion, Azamethyphos according to their genotoxic effects. The present study pointed out that the correct administration of pesdicides has significant effect on human and enviromental health which are under the threat of hazardous pesticid residues.
