Okaliptus ve bazı ticari uçucu yağlarının Domates bakteriyel kanser hastalığı (Clavibacter michiganensis subsp. michiganensis) üzerine antibakteriyel etkileri

Özet
Clavibacter michiganensis subsp. michiganensis karantinaya tabi, tohum kökenli, domates bitkisinde bakteriyel solgunluk ve kanser hastalığına sebep olan önemli Gram pozitif bir bakteriyel hastalık etmenidir. Bu çalışmanın amacı, okaliptus (Eucalyptus citriodora), sarı kantaron (Hypericum perforatum), karanfil (Dianthus caryophyllus), anason (Pimpinella anisum) ve hardal (Brassica nigra) bitki uçucu yağlarının in vitro ve in vivo koşullarda C. michiganensis subsp. michiganensis üzerine antibakteriyel etkinliğini belirlemektir. Bu amaçla, Mersin ilinden toplanan hastalıklı domates örneklerinden izole edilen bakteri izolatları morfolojik ve moleküler tanı yöntemleri ile incelenmiş, 6 tanesinin C. michiganensis subsp. michiganensis ile enfekteli olduğu teşhis edilmiştir. Uçucu yağların C. michiganensis subsp. michiganensis üzerine etkileri in vitro koşullarda disk difüzyon metodu ile araştırılmıştır. Test edilen uçucu yağlar arasında okaliptus uçucu yağının C. michiganensis subsp. michiganensis’in gelişimine en etkili yağ olduğu belirlendikten sonra, doz artışına bağlı antibakteriyel etkinliği belirlemek için antibiyogram disklerine farklı miktarlarda uçucu yağlar eklenerek engelleme zonları ölçülmüştür. Araştırma sonucunda, C. michiganensis subsp. michiganensis’e karşı okaliptus yağının en etkili dozunun 23 mm engelleme zonu oluşturan 25 μl yağ dozu olduğu tespit edilmiştir. Belirlenen uçucu yağ miktarı farklı kombinasyonlarla domates bitkisine uygulanmıştır. Bu sonuçlara göre, okaliptus uçucu yağının C. michiganensis subsp. michiganensis gelişimini engellediği tespit edilmiş ve mücadelesinde kullanılabileceği ortaya konulmuştur.
Abstract
Clavibacter michiganensis subsp. michiganensis is a Gram-positive, seed borne, and quarantine bacterium which causes bacterial wilt and cancer disease in tomato. This study was conducted to determine in vitro antibacterial activity of commercial plant essential oils of eucalyptus (Eucalyptus citriodora), St. John's wort (Hypericum perforatum), clove (Dianthus caryophyllus), anise seed (Pimpinella anisum) and mustard (Brassica nigra) on C. michiganensis subsp. michiganensis. In this study, morphological and molecular diagnostic assays were performed on suspicious samples collected from Mersin province and as a result six of them were identified as C. michiganensis subsp. michiganensis. The most effective essential oil on C. michiganensis subsp. michiganensis was determined by disc-diffusion method under in vitro conditions and the inhibition zone diameters were measured. Since eucalyptus essential oil was determined as the most efficient essential oil, the most effective concentration was also determined by applying different amount of essential oils on the antibiogram discs and measuring inhibition zones. The most effective dose was found to be 25 μl which caused 23 mm inhibition zone. This concentration was also applied on tomato plants in different combinations. Based on these results, it can be concluded that eucalyptus essential oils can be used for management of control strategies on C. michiganensis subsp. michiganensis.
