Detecting p53 exon 7 mutations of colorectal carcinomas by restriction enzyme analysis

Abstract
Wide variety of tumors proceed through an accumulation of genomic alterations involving activation of oncogenes and/or loss of tumor suppressor gene activities. Colorectal tumorigenesis is the model system for human tumor in which multistep changes occur in the course of development and progression of neoplasia. In this study, we aimed to investigate the codon 248 (exon 7) mutations in p53 tumor suppressor gene. Exon 7 mutations were detected in three of 17 primary tumor samples from colorectal cancer patients. The method of single strand conformation polymorphism was used by silver staining and the mutation in codon 248 was confirmed by Msp I restriction enzyme digestion analysis.
