Özet

Amaç: Non-sendromik yarık dudak/damak (nsYD/D) malformasyonu (CL/P, MIM 119530) en yaygın doğumsal defektlerden biridir. 19q13 dahil bazı kromozom bölgeleri (2p13,4p,4q,6p ve 19q13) ve bu bölgelerde yer alan BCL3, PVR, PVRL2 and CLPTM1 gibi birçok gen allelik ilişkilendirme ile ilgili sendromda rol oynayıp oynamadığını test etmek için sıkça çalışılmıştır. Bunlardan CLPTM1 geni ilk olarak 2 veya 3 nesil boyunca segrege olan 19q, 2q translokasyon taşıyıcısı yarık dudak/damaklı bir ailede belirlenmiştir. Gereç ve Yöntemler: Bu çalışmada 19q13 kromozom bölgesinde yer alan CLPTM1 geninin nsYD/D malformasyonu ile ilişkisinin olup olmadığı araştırılmıştır. Bunun için, Guatemala’dan toplam 79 olgu ve 57 kontrol grubunda CLPTM1 geni 3. ekzonunda SSCP tekniği ile mutasyon taraması yapılmıştır. Sonuç olarak, CLPTM1 geninin 3. ekzonunda 2 adet sessiz mutasyon, A88A and P85P, saptanmış ve takiben de A88A varyantının nsYD/D’deki rolünü test etmek için mutant alel frekansları hesaplanarak olgu ve kontrol grubunda farklı olup olmadığı incelenmiştir. Bulgular: İncelenen T aleli bakımından olgularla kontrol bireyler arasında bir fark gözlenmemiştir. Böylece, çalışılan Guatemala populasyonunda CLPTM1 geni A88A polimorfizmi ile nsYD/D malformasyonu arasında bir ilişki bulunamamıştır. Sonuç: Bu bulgular CLPTM1 geninin nsYD/D malformasyonundaki rolünü, en azından çalışılan bu populasyonda, desteklememektedir.

Abstract

Purpose: Non-syndromic cleft lip with or without cleft palate (CL/P, MIM 119530) is perhaps one of the most common major birth defects. Many chromosomal regions including 19q13 (2p13,4p,4q,6p and 19q13) and many candidate genes such as BCL3, PVR, PVRL2 and CLPTM1 were analyzed through allelic association studies to test their involvement in non-syndromic cleft lip and palate. The latter one was originally identified in a family with 19q, 2q translocation segregated two or three generation. Methods: In this study, CLPTM1 gene localized to 19q13 region were analyzed for its association with the non-syndromic CL/P malformation. For this, a total of 79 patients with non-syndromic CL/P and 57 healthy controls from Guatemala were screened for variations at exon 3 using PZR-SSCP technique. As a result, two synonymous changes were determined as A88A and P85P in the CLPTM1 gene followed by calculation of mutant allele frequency of the variant, A88A, to test its association with nsCL/P. Results: The CLPTM1 T allele frequency was not found to be different in patients versus controls. In conclusion, no association was observed between the A88A polymorphism in the CLPTM1 gene and non-syndromic cleft lip with/without palate malformation in the Guatemalan population. Conclusion: These data suggest that CLPTM1 gene does not participate in the development of nsCL/P, at least, in the Guatemalan population studied.
