Protective effect of Nigella sativa on lung injury induced by chronic toluene exposure in rats

Özet
Amaç: Bu çalışma, kronik toluen maruziyetinden sonra sıçanların akciğerlerinde oluşacak hasara karşı Nigella sativa’nın (NS) muhtemel koruyucu etkilerini araştırmak amacıyla planlandı. Gereç ve Yöntem: Sıçanlar, herbir grup 10 hayvan içerecek şekilde biri kontrol ikisi deney olmak üzere rastgele 3 gruba ayrıldı; Kontrol grubu, toluen uygulanan grup ve toluen+NS uygulanan grup. Toluen, 12 hafta boyunca, haftada 6 gün ve günde 8 saat süre ile 3000 ppm toluen inhalasyon yolu ile uygulanmıştır. NS uygulanan gruptaki ratlara, 400 mg/kg NS 12 hafta boyunca ağızdan günde bir kez, toluene uygulanmasından hemen sonra verilmeye başlandı. Kontrol grubuna da 1 ml serum fizyolojik uygulandı. Doku örnekleri histopatolojik incelemeler için alındı. Son zamanlarda, toluene maruz kalmış sıçanların akciğerlerindeki histopatolojik değişikliklerin NS tedavisi ile ilgili herhangi bir çalışma rapor edilmemiştir. Bulgular: Çalışmamız toluen uygulanan sıçanlarda NS’nin pulmonar inflamatuar cevabı inhibe ettiği, peribronşiyal hücre infiltrasyonu, alveolar septal infiltrasyonu, alveolar eksudatı, interstitial fibrozisi ve nekrozu anlamlı bir şekilde azalttığı gösterilmiştir. Bulgularımız, NS ile birlikte toluen uygulanan akciğerlerde apoptotik aktivitenin anlamlı bir şekilde azaldığını göstermiştir. Sonuç: NS’nın faydası konusunda ileride preklinik araştırmaların, toluene maruz bırakılan sıçanlarda akciğer hasarına karşı potansiyel bir tedavi olarak faydalı olabileceğine inanıyoruz
Abstract
Purpose: The aim of this study was designed to evaluate the possible protective effects of Nigella sativa (NS) on the lung injury in rats after chronic toluene exposure. Methods: The rats were randomly allotted into one of three experimental groups: control, toluene treated and toluene treated with NS; each group contain 10 animals. Control group received 1ml serum physiologic and toluene treatment was performed by inhalation of 3000 ppm toluene, in a 8 hr/day and 6 day/week order for 12 weeks. The rats in NS treated group was given NS (in a dose of 400 mg/kg body weight) once a day orally for 12 weeks starting just after toluene exposure. Tissue samples were obtained for histopathological investigation. To date, no histopathological changes of lung in rats after chronic toluene exposure by NS treatment have been reported. Results: Our study showed that NS treatment inhibits the inflammatory pulmonary responses reducing significantly peribronchial inflammatory cell infiltration, alveolar septal infiltration, alveolar edema, alveolar exudate, interstitial fibrosis, and necrosis formation in toluene treated rats. Our data indicate a significant reduction in the activity of in situ identification of apoptosis using terminal dUTP nick end-labeling (TUNEL) in lung tissue of toluene treated with NS therapy. Conclusion: We believe that further preclinical research into the utility of NS may indicate its usefulness as a potential treatment on lung injury after chronic toluene exposure in rats.
