Özet

Hücre siklüsünde geç Gj ve S fazlarında sentezlenen ve DNA replikasyon proteini olan Prolifera¬ting Cell Nuclear antigen (PCNA), anti-PCNA monoklonal antikorların varlığı ile tümör hücrelerinin prolife-rasyon hızını göstermede kullanılmaya başlanmıştır. Bu çalışmada, squamous hücreli larenks karsinomu tanısı konulmuş 20 olguda, immunohistokimyasal olarak PCNA değerleri araştırıldı. Olgular düşük PCNA (median'ın altındaki değerler) ve yüksek PCNA (median'ın üstündeki değerler) pozitifliğine sahip olanlar olarak 2 gruba ayrıldı. PCNA değerleri yüksek olan gruptaki olgularda, yüksek oranda lenf metastazı tespit edildi Sürvi analizi yapıldığında 2 grup ara¬sında anlamlı bir fark bulunurken, histolojik dijferansiasyon ve PCNA arasında çok anlamlı bir ilişki tes¬pit edilmedi (ancak sonuç anlamlılık sınırında kabul edildi). Tüm bu sonuçlar, PCNA'in squamous hücreli larenks karşinomlannda, prognozu belirlemede faydalı bir biomarker olduğunu göstermiştir.

Abstract

Proliferating Cell Nuclear Antigen (PCNA) is a DNA replication protein, maximally elevated in late Gj and S phases of the cell cycle. By using monoclonal antibodies, the expression of PCNA can be quantified and the rate of tomur cell proliferation estimated. In this study. PCNA expression was investigated immunhistochemically in 20 squamous cell carcinomas of the larynx. By dividing the patients into groups of low PCNA positivity (less than the median) and high PCNA positivity (greater than the median), it was found that there was a strong trend towards a higher degree of nodal involvement with increasing PCNA positivity. A survival analysis was also performed, significant differences could be found between the two groups. But there was no significant differences was found to histological differentiation. These results indicate that PCNA could be a useful biomarker as a prognostic factor in patients with squ¬amous cell carcinoma of larynx.
